宫颈脱落细胞中人乳头瘤病毒52型感染及整合状态分析

摘要 目的探讨人乳头瘤病毒(Human papillomavirus,HPV)感染及其整合状况在宫颈病变中的意义。方法应用HPV L1通用引物MY09/11和HPV52 E6型特异性引物进行多聚酶链反应(Polymerase chain reaction,PCR)检测宫颈脱落细胞标本中HPV总感染及HPV52感染状况;应用荧光定量PCR检测HPV52 E6、E2及人管家基因β-actin的拷贝数,通过E2与E6比值分析HPV52整合状况。结果928例宫颈脱落细胞标本中检测到219例HPV阳性。检测到26例HPV52阳性,其中5例为纯游离型,即E2与E6比值等于1;14例为整合与游离的混合型,即E2/E6比值在0与1之间;7例为纯整合型,即E2/E6比值等于零。结论HPV52整合入宿主基因组可能是宫颈癌变的关键因素;通过HPV52感染及整合状况分析,对宫颈病变患者的早期筛查具有一定意义。 To investigate the importance of human papillomavirus(HPV)infection and integration,HPV and HPV52 positive cervical lesions were detected in cervical exfoliated cells through MY09/11 polymerase chain reaction(PCR)by using HPV L1 primers and HPV52 E6 specific primers.The number of copies of HPV52 E6,E2 and the human housekeeping geneβ-actin was detected with fluorescence quantitative PCR,and the HPV52 integration status was analyzed according to the ratio of E2 to E6.A total of 219 samples from 928 cases of cervical exfoliated cells were HPV positive,and 26 samples from 219 HPV positive cases were HPV52 positive.Among 26 HPV52 positive samples,there were 5 pure episomal cases with E2/E6 ratios=1;14 mixed integrated and episomal cases with an E2/E6 ratio of 0 to 1;and 7 pure integrated cases with an E2/E6 ratio=0.HPV52 integration into the host genome may therefore be a key factor in cervical lesions.The analysis of HPV52 infection and integration status has a certain significance for early screening of patients with cervical lesions.
