摘要
目的:探讨卵巢上皮性癌OVCAR-3和OVCAR-8细胞中miR-338-3p过表达对MET转录调节因子(MACC1)表达及细胞迁移和侵袭的影响。方法:在线医学数据库检索miR-338-3p在卵巢上皮性癌组织中的表达,预测其下游靶基因,双荧光素酶报告实验验证其靶基因;miR-338-3p和MACC1过表达慢病毒载体及阴性对照慢病毒载体分别转染OVCAR-3和OVCAR-8细胞,Transwell实验分析其对OVCAR-3和OVCAR-8迁移和侵袭的影响,Western blot法检测经典Wnt/β-catenin信号通路相关蛋白表达的影响。结果:miR-338-3p在上皮性卵巢癌组织中的表达低于正常卵巢组织,MACC1是miR-338-3p下游的靶基因之一。过表达MACC1可促进OVCAR-3和OVCAR-8细胞的迁移和侵袭(P<0.05),而过表达miR-338-3p可抑制OVCAR-3和OVCAR-8细胞的迁移和侵袭(P<0.05),也可抑制MACC1、Wnt3a和p-LRP6蛋白的表达(P<0.05)。结论:miR-338-3p可能通过调节MACC1和经典Wnt/β-catenin通路影响卵巢上皮性癌细胞的迁移和侵袭。
Aim:To explore the effects of miR-338-3p overexpression in epithelial ovarian cancer OVCAR-3 and OVCAR-8 cells on MACC1 expression and cell migration and invasion.Methods:Multiply biomedical databases were queried for the expression of miR-338-3p in epithelial ovarian cancer tissue and downstream target gene,double luciferase report experiment was used to verify its target gene.Overexpressing miR-338-3p and MACC1 lentiviral vectors were transfected into OVCAR-3 and OVCAR-8 cells,cell migration and invasion were analyzed by Transwell assay.Canonical Wnt/β-catenin pathway related proteins were measured by Western blot.Results:The expression of miR-338-3p was decreased in epithelial ovarian cancer tissue compared with normal ovarian tissue.MACC1 is one of downstream target genes of miR-338-3p.Overexpression of MACC1 could induce the migration and invasion of OVCAR-3 and OVCAR-8 cells(P<0.05).Overexpression of miR-338-3p could suppress the migration and invasion of OVCAR-3 and OVCAR-8 cells(P<0.05),and inhibit the expressions of MACC1,Wnt3a and p-LRP6(P<0.05).Conclusion:miR-338-3p may be involved in the migration and invasion of epithelial ovarian cancer cells,which might via the regulation of MACC1 and canonical Wnt/β-catenin signaling pathway.
