摘要 基于可视化免疫亲和凝胶柱(IATC)方法,建立了一种快捷、准确的检测牛奶和肉类中链霉素残留的方法.整个检测过程在固相萃取(SPE)柱中进行,柱中包含一个结合链霉素多克隆抗体的检测层和一个结合辣根过氧化物酶(HRP)抗体的质控层.将样品处理液与酶标抗原预混合后加入到凝胶柱中,根据凝胶柱中检测层颜色的变化快速筛查牛奶和肉类中链霉素的残留.整个检测过程仅需10 min完成,检出限为10μg/kg.可视化IATC法对牛奶中链霉素的检出限为40μg/kg,对牛肉和猪肉中链霉素的检出限均为160μg/kg.取牛奶、牛肉、猪肉共12件空白样品制成盲样用于检测,其结果与高效液相色谱-串联质谱(HPLC-MS)一致. A fast and accurate method for the detection of streptomycin(STR)residues in milk and meat was established based on the visual immunoaffinity gel column(IATC)method.The whole detection process was carried out in a solid phase extraction(SPE)column,which comprised a detection layer binding streptomycin polyclonal antibody and a quality control layer binding horseradish peroxidase(HRP)antibody.The sample treatment solution was premixed with enzyme-labeled anti-gen and then added into a gel column,and streptomycin residues in milk and meat were rapidly screened according to the color change of the detection layer in the gel column.The whole detection process can be completed in only 10 min,reaching the requirement of rapid detection.The detection limit of the gel column was 10μg/kg.The detection limit of STR was 40μg/kg in milk and 160μg/kg in beef and pork by IATC method.A total of 12 blank samples of milk,beef and pork were taken and made into blind samples for detection.The results were consistent with those of high performance liquid chromatographytan-dem mass spectrometry(HPLC-MS).
