摘要
目的:探讨父子二联体亲子鉴定中STR出现两步突变基因座的原因。方法:应用多重PCR扩增技术及毛细管电泳技术进行基因座分型,本DNA进行检测分析。结果:常染色体STR检测结果中Penta E基因座存在对样两步突变,其他STR基因座检测结果均符合遗传规律。结论:亲权鉴定过程中,若发现基因座出现多步突变,应增加STR位点计算累计亲权指数综合分析来排除或支持亲子关系。
Objective:To investigate the causes of the occurrence of two-step mutant loci on STR in paternity testing.Methods:Multisite PCR and capillary electrophoresis were used to classify the loci and analyze the sample DNA.Results:Two-step mutation of Penta E locus was found in the autosomal testing system,and the results of other STR loci were consistent with the genetic law.Conclusion:If multi-step mutation of locus is found in paternity identification,comprehensive analysis of genetic markers should be added to exclude or identify parent-child relationship.
