PRRSV/LPS共刺激抑制小鼠雌二醇分泌与苦参碱的改善作用

摘要 本试验旨在探讨PRRSV/LPS共刺激对小鼠雌二醇(E2)分泌的影响,以及探究苦参碱对共刺激引起E2变化的影响。试验分为空白组,PRRSV与LPS共刺激组和苦参碱治疗组。给药7 d后采集血液,ELISA法测定血清E2水平;冻存卵巢组织,qRT-PCR检测StaR、CYP11a1、CYP19a1、3β-HSD、17β-HSD、Caspase-3、Bax和Bcl-2 mRNA的表达。结果表明,与空白组相比,PRRSV与LPS共刺激组中血清E2含量降低(P<0.05),且编码E2合成过程中关键酶的基因(StaR、CYP11a1、CYP19a1)mRNA表达显著降低(P<0.05),凋亡相关基因(Caspase-3、Bax、Bcl-2)mRNA表达无显著变化(P>0.05)。苦参碱可以明显升高由PRRSV/LPS共刺激诱导的E2含量降低,显著升高CYP11a1 mRNA的表达(P<0.05)。结果提示,PRRSV/LPS共刺激降低小鼠E2分泌,而苦参碱可以抑制共刺激诱导的E2分泌降低。 The aim of study was to investigate the effect of PRRSV and LPS co-stimulation on the secretion of 17β-estradiol(E2)in mice.At the same time,we explored the effect of matrine on hormonal changes caused by co-treatment.The experiment was divided into control group,PRRSV and LPS co-stimulation group and matrine group.Blood samples were collected at 7 days after administration,E2 level in serum was determined by ELISA.Ovarian tissue was frozen,and qRT-PCR was used to determine StaR,CYP11a1,CYP19a1,3β-HSD,17β-HSD,caspase-3,Bax,Bcl-2 mRNA expression levels.The results showed that compared with the control group,PRRSV/LPS co-stimulation group significantly decreased the serum E2 content(P<0.05)and gene mRNA expression(StaR,CYP11a1,CYP19a1)encoding key enzymes during the synthesis(P<0.05),but not significant for apoptosis-related genes(P>0.05).Matrine significantly increased the decrease of E2 content induce by PRRSV/LPS co-stimulation and the expression of CYP11a1 mRNA(P<0.05).These results indicates that PRRSV/LPS co-stimulation reduces E2 secretion in mice,while matrine inhibits E2 reduction.
