胆畅心宽茶对小鼠胆囊胆固醇结石形成影响的实验研究

摘要 目的:探究胆畅心宽茶对小鼠胆囊胆固醇结石形成影响及潜在机制。方法:给予小鼠普通饲料+/-胆畅心宽茶以及成石饲料+/-胆畅心宽茶,每组9只。喂养4周后处死采集血清、胆囊胆汁、肝脏等标本做检测。结果:单纯成石饲料组小鼠,胆囊内均出现肉眼结石以及镜下胆固醇结晶100%,而成石饲料加茶组肉眼结石只有44%,镜下胆固醇结晶78%。在普通饲料及成石饲料、饮茶小鼠胆汁中β-鼠胆酸比例均显著增高。成石饲料加茶组,肝脏胆汁酸代谢相关基因Cyp8b1表达下降,Cyp2c70表达上升。结论:胆畅心宽茶能预防胆囊胆固醇结晶/结石形成,可能与促进β-鼠胆酸合成,提高胆汁中胆汁酸亲水性有关。 Objective:To explore the potential mechanism and the effect of Danchangxinkuan tea on the formation of cholesterol gallstones in mice.Methods:Nine mice in each group were given chow diet+/-tea or lithogenic diet+/-tea.After 4 weeks of feeding,mice were sacrificed to collect serum,gallbladder bile,liver and other specimens for testing.Results:Gross stones and cholesterol crystals found in the gallbladder of all mice accounted for 100%in the lithogenic diet group when observed by naked eyes or under the microscope,only 44%(by naked eyes)and 78%(under the microscope)in the lithogenic diet plus tea group.The ratio ofβ-murocholic acid in the bile of tea-drinking mice was significantly increased in both diets.The expression of liver bile acid metabolism-related gene Cyp8b1 decreased while the expression of Cyp2c70 increased in lithogenic diet plus tea group.Conclusion:Danchangxinkuan tea can prevent gallbladder cholesterol crystals/stone formation,which may be related to the promotion ofβ-murocholic acid synthesis and the improvement of hydrophilicity of cholic acid in bile.
