摘要 目的优化黄芩中黄芩苷的最佳水提工艺。方法采用单因素结合正交试验设计法,以黄芩苷提取率和含量为评价指标,对影响黄芩苷制备工艺的影响因素(浸泡条件、溶剂用量、提取时间、提取次数)进行考察。结果最佳提取工艺为10+5倍药材量的沸水,提取2次,时间分别为60 min和30 min,抽滤,合并两次提取液,用4 mol/L盐酸调节pH值至1~2,80℃水浴30 min,静置12 h,离心,水洗至中性,40℃干燥即得。在此条件下,黄芩苷提取率15.08%,纯度84.58%。结论该制备工艺操作简便、稳定,黄芩苷提取率和纯度较高,且成本低,可以为工业大规模生产提供理论依据。 Objective To optimize the water extraction process of baicalin from Scutellaria baicalensis.Methods The extraction rate and content of baicalin were used as evaluation indexes to investigate the influencing factors(soaking conditions,solvent dosage,extraction time and extraction times)of baicalin preparation process by single factor test combined with orthogonal design.Results The best extraction process was:ten plus five folds of boiling water,extracting twice,for 60 min and 30 min respectively.After filtration,the extracts were mixed together,and then the pH of the filtrate was adjusted to 1 to 2 with hydrochloric acid.The solution was kept in a water bath at the temperature of 80℃for 30 minutes,then laid for 12 h to deposit completely and filter.The deposit was washed with water till the pH value was neutral and then heated at 40℃for drying to obtain crude baicalin.The extraction rate and purity of baicalin were 15.08%and 84.58%under this condition.Conclusion The extraction process is stable and feasible with high extraction rate,high purity,and low cost,which can provide a theoretical basis for the industrial large-scale production of baicalin.
