摘要
目的基于生物信息学分析人STN1蛋白的结构、功能。方法从Uniprot数据库中获取人STN1蛋白氨基酸序列,使用ProtParam在线工具分析其理化性质,DNAMAN软件分析其亲/疏水性,通过Genecar网站查看其在细胞中的分布。分别通过PSIPRED网站、PDB数据库、Uniprot网站和NCBI Conserved Domain数据库预测人STN1蛋白的二级结构、三级结构、结构域及保守结构域。通过NCBI网站及DNAMAN软件对人STN1蛋白进行多重序列比对,通过MEGA软件构建系统进化树。利用GPS5.0软件预测人STN1蛋白的磷酸化及乙酰化位点,使用AmiGO2网站对人STN1蛋白进行基因本体(GO)分析,Reactome在线工具查看其参与的信号通路。通过STRING数据库预测人STN1蛋白的相互作用蛋白,K-Means算法对相互作用蛋白进行聚类分析。结果人STN1蛋白由368个氨基酸组成,是一种偏酸性、不稳定、疏水的胞内蛋白,在进化过程中高度保守。其由一个OB结构域和两个wHTH基序组成,其中wHTH区域为人STN1蛋白的保守区;OB结构域涵盖了大部分(77.8%)的磷酸化位点。GO分析显示,在分子功能方面,人STN1蛋白主要具有与蛋白质、DNA结合的功能;在生物学途径方面,人STN1蛋白对端粒酶具有负调控作用,也是DNA复制的正调控因子。人STN1蛋白除可与TEN1、CTC1形成复合物外,还可与8个介体复合物亚基直接结合。结论人STN1蛋白为偏酸性、不稳定、疏水的胞内蛋白,其组成包括一个OB结构域和两个wHTH基序,是参与基因组复制调控和维持端粒稳态的重要蛋白。
Objective To analyze the structure and function of human STN1 protein based on bioinformatics.Meth⁃ods We obtained the amino acid sequence of human STN1 protein from Uniprot database,used ProtParam online tool to analyze its physical and chemical properties,DNAMAN software to analyze its affinity/hydrophobicity,and checked its distribution in cells through the Genecar website.The secondary structure,tertiary structure,domain and conserved domain of human STN1 protein were predicted by PSIPRED,PDB,Uniprot and NCBI Conserved Domain,respectively.Multiple sequence alignments of human STN1 protein were carried out through NCBI website and DNAMAN software,and a phylogenetic tree was constructed through MEGA software.We employed GPS5.0 software to predict the phosphorylation and acetylation sites of human STN1 protein,used the AmiGO2 to perform gene ontology(GO)analysis on human STN1 protein,and employed the Reactome online tool to view the signal pathways which they participated in.The interacting proteins of human STN1 protein were predicted by STRING database,and the K-Means algorithm was used to cluster the interacting proteins.Results The human STN1 protein was composed of 368 amino acids.It was an acidic,unstable,and hydrophobic intracellular protein that was highly conserved during evolution.It consisted of an OB domain and two wHTH motifs,where the wHTH region was a conserved region of human STN1 protein;the OB domain covered most(77.8%)phosphorylation sites.GO analysis showed that in terms of molecular function,human STN1 protein mainly had the function of binding to protein and DNA;in terms of biological pathways,human STN1 protein had a negative regulatory effect on telomerase and was also a positive regulator of DNA replication.In addition to forming a complex with TEN1 and CTC1,human STN1 protein could also directly bind to eight mediator complex subunits.Conclusions The human STN1 protein is an acidic,unstable,and hydrophobic intracellular protein.It includes an OB domain and two wHTH motifs.It is an important protein involved in the regulation of genome replication and maintains the telomere homeostasis.
