基于数据挖掘分析Elafin在肺癌中的表达及意义

摘要 目的通过数据库数据分析肺癌中弹性蛋白酶抑制因子(Elafin)的表达及意义。方法Oncomine数据库分析Elafin基因在肺癌中表达情况并通过实时荧光定量PCR(qPCR)进行验证;Kaplan-Meier数据库分析Elafin表达与肺癌患者预后的关系;Linkedomics数据库分析Elafin基因在TCGA LUAD中的相关基因;通过GSEA tools对Elafin的相关基因进行基因本体论(GO)功能注释、京都基因与基因组百科全书(KEGG)通路分析;string-db数据库构建Elafin蛋白相互作用网络。结果Oncomine及qPCR结果显示,肺癌组织中Elafin的mRNA表达水平较正常组织显著增高;Kaplan-Meier生存分析显示,Elafin高表达患者总生存期和无病生存期较低表达者明显降低;Linkedomics分析显示Elafin有7697个相关基因;GO分析、KEGG分析显示,Elafin及其相关基因可能与体液免疫反应、粒细胞活化、白细胞介素(IL)-17信号通路、核因子(NF)-κB信号通路等相关;Elafin蛋白相互作用网络显示Elafin与Loricrin角化包膜前体蛋白(LOR)、胱抑素A(CSTA)、丝蛋白(FLG)等相互作用。结论Elafin在肺癌中表达明显增高且与患者预后相关,是肺癌潜在的诊断与治疗靶标。 Objective To investigate the expression and significance of Elafin in lung cancer by database through database data.Methods Elafin gene expression in lung cancer was analyzed by Oncomine database and verified by realtime fluorescence quantitative PCR(qPCR)in lung cancer tissues.Kaplan-Meier database was used to analyze the relationship between Elafin expression and the prognosis of lung cancer patients.The correlated genes of Ealfin were obtained from Linkedomics database.GO and KEGG analysis and protein-protein interaction network were analyzed by GSEA tools and string databases,respectively.Results Compared with normal tissue,the mRNA level of Elafin was significantly higher in lung cancer.Survival analysis showed that disease-free survival and overall survival were significantly decreased in patients with high expression of Elafin.Linkedomics database showed that there were 7697 correlated genes in Elafin.GO and KEGG analysis showed that Elafin and correlated genes may related to humoral immune response,granulocyte activation,IL-17 pathway and NF-κB pathway.The Elafin protein PPI network showed that Elafin may interact with LOR,CSTA and FLG.Conclusion The high expressed Elafin in lung cancer tissues is associated with the prognosis of lung cancer patients,which may be used as a potential target for diagnosis and treatment of lung cancer.
