摘要
为了获得泰妙菌素高产菌株,以泰妙菌素19-127为出发菌株,采用微波对泰妙菌株进行诱变,并结合特比萘酚及制霉菌素2种抗生素经过抗性筛选法获得泰妙菌素高产菌株。结果表明,经过发酵验证最终选育出5株突变株,分别为20-016、20-029、20-048、20-072和20-088,较对照组效价分别增长19.9%、21.6%、20.2%、22.5%和23.1%,且连续传代5次,遗传性状稳定。比较2种抗生素在泰妙菌素高产菌株选育中的作用,制霉菌素更有利于高产菌株的选育。
In order to obtain tiamulin high-yield strain,tiamulin 19-127 was used as the starting strain,and the tiamulin strain was mutagenized by microwave,and the tiamulin high-yield strain was obtained through resistance screening method on the basis of terbinacol and nystatin.The results showed that 5 mutant strains were selected after fermentation verification,namely 20-016,20-029,20-048,20-072 and 20-088.Compared with the control group,the titer increased by 19.9%,21.6%,20.2%,22.5%and 23.1%,respectively,and the genetic characters were stable after 5 successive generations.Comparing the effects of the two antibiotics in the breeding of tiamulin high-yield strains,nystatin was more beneficial for the breeding of high-yield strains.
