摘要
目的:探讨祛风解痉方对哮喘模型小鼠γδT淋巴细胞表达的影响。方法:以卵清蛋白滴鼻的方法建立哮喘Balb/c小鼠模型。动物分组:将30只小鼠随机分为正常组、模型组、醋酸泼尼松片组(0.27 mg/d)及3个祛风解痉方组(0.59、1.18、2.36 g/d),分别进行干预,给药14 d,1次/d。结果:与哮喘模型组比较,祛风解痉方低剂量组的γδT细胞的细胞活性差异无统计学意义(P>0.05),而祛风解痉方中剂量组、祛风解痉方高剂量组及阳性药物组的γδT细胞的细胞活性显著降低(P<0.05)。与模型对照组比较,祛风解痉方低剂量组的IL-5、IL-13、TGF-β、IL-10、IFN-γ水平差异无统计学意义,而祛风解痉方中剂量组、高剂量组以及醋酸泼尼松片组在治疗后IL-5、IL-13、TGF-β的表达均显著降低(P<0.05),IL-10、IFN-γ的表达均显著升高(P<0.05)。结论:祛风解痉方通过抑制γδT细胞的细胞活性,降低γδT细胞分泌的细胞因子IL-5、IL-13、TGF-β表达,调高IL-10、IFN-γ的表达抑制气道炎性反应,改善哮喘小鼠的气道高反应性。
Objective:To investigate the effects of Qufeng Jiejing Formula on the expression ofγδT lymphocyte in asthmatic model mice.Methods:The asthmatic Balb/c mice model was established by ovalbumin nasal drops.Animal grouping:a total of 30 experimental mice were randomly divided into a normal group,a model group,a prednisone acetate tablet group(0.27 mg/d)and a 3 Qufeng Jiejing Formula groups(0.59,1.18,2.36 g/d).The mice were given intervention for 14 days,once a day.Results:Compared with the asthma model group,there was no significant difference in the cell activity ofγδT cells in the low-dose Qufeng Jiejing Decoction group(P>0.05),while the middle-dose Qufeng Jiejing Decoction group and the high-dose Qufeng Jiejing Decoction group and the cell viability ofγδT cells in the positive drug group was significantly reduced(P<0.05).Compared with the model control group,the levels of IL-5,IL-13,TGF-β,IL-10 and IFN-γin the low-dose group of Qufeng Jiejing Formula had no significant difference,while the levels of IL-5,IL-13 and TGF-βin the middle-dose group,the high-dose group and the prednisone acetate tablet group decreased significantly after treatment(P<0.05),while the levels of IL-10 and IFN-γincreased significantly(P<0.05).Conclusion:Qufeng Jiejing Formula can by inhibiting the cell activity ofγδT cells,reduce the expression of cytokines IL-5,IL-13,and TGF-βsecreted byγδT cells,increase the expression of IL-10 and IFN-γ,inhibit airway inflammation,and improve asthmatic mice.
