不同大麦品种特异性SSR分子标记的筛选 被引量:7

摘要 为探究鉴别大麦品种特异性所需要SSR标记的数量与鉴别效果,以国内外44个大麦品种为材料,并根据来源不同,将其分为江浙沪组、国内组和国际组;根据SSR标记的多态性及其分布,选取位于大麦7条染色体上的28个SSR标记对参试品种进行遗传多样性分析与特异性鉴定,研究鉴别不同来源组内不同品种所需的最少标记数和最佳标记数。结果表明,鉴别江浙沪组、国内组和国际组大麦品种所需要的最少SSR标记数分别为7、6和6个。考虑到分子标记在大麦染色体上分布的均匀性及其对同一来源组其他大麦品种鉴别的广适性,并结合各分子标记的多态性及不同分子标记组合的品种聚类效果,初步认为鉴别江浙沪组、国内组和国际组所需要的最佳SSR标记数分别为14、15和15个,说明15个SSR标记组合可以鉴别不同来源大麦品种的真实性和纯度。 In order to explore the quantity and efficiency of SSR markers for identification of barley variety distinctness,44 domestic and international barley varieties have been used as materials,which were divided into three groups according to their sources:Jiangsu-Zhejiang-Shanghai group,domestic group and international group.According to the polymorphism and distribution of SSR markers,28 SSR markers located on 7 chromosomes of barley were selected for genetic diversity analysis and distinctness identification of the tested varieties,so as to determine the minimum number and the optimal number of the markers for identification of different varieties in different groups.The results showed that the minimum number of SSR markers for identifying barley varieties in Jiangsu-Zhejiang-Shanghai group,domestic group and international group were 7,6 and 6,respectively.Considering the distribution uniformity of the molecular markers on barley chromosome and its wide adaptability for identification of other barley varieties in the same group,combined with the polymorphism of each molecular marker and the clustering effect of different combinations of molecular markers,it is preliminarily considered that the optimal number of SSR markers for distinguishing Jiangsu-Zhejiang-Shanghai group,domestic group and international group are 14,15 and 15,respectively.In general,we can identify the distictness of barley varieties from different sources by 15 SSR markers.
