基于中药整合药理学平台的参桃软肝方抗癌分子机制研究

Molecular Anti-Cancer Mechanism Research on Shentao Ruangan Recipe Based on Integrative Pharmacology

目的:基于中药整合药理学平台筛选参桃软肝方抗癌治疗的主要成分及作用靶点,构建相关网络对参桃软肝方的抗癌机制进行预测分析。方法:借助中药整合药理学平台及DAVID数据库对参桃软肝方行整合药理学分析,从多角度探索其抗癌机制。结果:研究结果表明,参桃软肝方中具有抗癌作用的药物成分种类丰富,抗癌治疗的关键靶点主要包括ADORA3、RHOA、AKT1、KRAS。涉及的关键通路包括T细胞受体信号通路、癌症通路及趋化因子信号通路,涉及的参桃软肝方潜在治疗癌种包括结直肠癌、胰腺癌、慢性髓性白血病等。结论:参桃软肝方具有多成分、多靶点干预肿瘤疾病的特点。本研究结果初步阐明参桃软肝方抗癌治疗的药效成分、作用靶点、分子通路及潜在的治疗癌种,为进一步深入揭示其作用机制奠定了基础。

Objective:To further investigate the potential molecular mechanism and active components of Shentao Ruangan recipe for treating cancer.Putative targets of Shentao Ruangan recipe were predicted and herbs-ingredients interaction network,ingredient-targets interaction network were constructed based on integrative pharmacology.Method:The traditional Chinese medicine of Integrative pharmacology database and DAVID database were applied to preform integrative pharmacology analysis of Shentao Ruangan recipe.The underlying mechanisms of Shentao Ruangan recipe for cancer treatment were analysed from multi-angles.Result:This study found that multiple ingredients of Shentao Ruangan recipe could exert therapeutic effect on cancer.The anti-cancer effect of Shentao Ruangan recipe was mainly associated with the hub targets including:ADORA3,RHOA,AKT1,and KRAS.The hub pathways involved were T cell receptor signaling pathway,pathways in cancer,and chemokine signaling pathway.In addition,Shentao Ruangan recipe might exert therapeutic effects on colorectal cancer,pancreatic cancer,chronic myeloid leukemia and other cancers.Conclusion:Shentao Ruangan recipe could treat cancer through multiple ingredients and multiple targets.The present study preliminarily revealed the active ingredients,hub targets,molecular pathways and potentially curable cancer types of Shentao Ruangan recipe,which will contribute to the further researches of this effective formula.
