摘要
目的:基于数据挖掘及网络药理学方法,探究中药治疗不孕症方剂的核心药组,分析其治疗不孕症的分子作用机制。方法:将收集到的不孕症方剂录入中医传承辅助系统(V2.5)获得核心药组,应用中药系统药理数据库和分析平台(TCMSP)获取核心药组的化学成分,根据ADME性质进行有效化学成分筛选并得到相应的靶点蛋白;以UniProt数据库为依据,获取靶点蛋白的基因名,对核心药组全部基因集进行GO、Kegg富集分析,构建核心药组关键靶基因集的蛋白互作网络,并对该网络基因集进行蛋白组织表达鉴定。结果:当归、香附、川芎等9味中药构成不孕症方剂核心药组,其靶基因主要参与信号传导、药物反应、细胞增殖的正调控等生物过程,并通过雌激素、催乳素、肾素分泌等信号通路,在子宫、肾、垂体等部位对不孕症发挥治疗作用。结论:阐明中医药治疗不孕症方剂的核心药组,揭示核心药组作用靶标参与的基因功能和相关信号通路,为深入研究中医药治疗不孕症的机制提供了理论依据。
Objective:To explore the core group of traditional Chinese medicine prescription for infertility and analyze the possible mechanism of its treatment of infertility based on data mining and network pharmacology methods.Methods:The collected prescription for infertility was recorded into the traditional Chinese medicine inheritance assistance system(V2.5)to obtain the core drug group;Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)were applied to obtain the chemical components of the core drug group,and the effective chemical components were screened according to the properties of ADME and the corresponding target proteins were obtained.Based on UniProt database,gene names of target proteins were obtained and all gene sets of core drug groups were analyzed by GO and Kegg enrichment.The protein interaction network of the key target gene set of the core drug group was constructed and the protein tissue expression of the network gene set was identified.Results:Angelica sinensis,Rhizoma cyperi,rhizoma chuanxiong and other 9 Chinese herbal medicines constitute the core drug group of infertility formula.Core drug group target genes are mainly involved in signal transduction,response to drug,positive regulation of cell proliferation and other biological processes.Through the estrogen,prolactin,renin secretion and other signaling pathways,in the uterus,kidney,pituitary and other parts of infertility play a therapeutic role.Conclusion:This paper elucidate the core group of traditional Chinese medicine prescription for infertility,and preliminarily reveal the gene function and related signal pathway involved in the target of the core group,which provides a theoretical basis for the in-depth study of the potential mechanism of traditional Chinese medicine treatment for infertility.
