基于新疆紫草中β,β′-二甲基丙烯酰阿卡宁含量变化的浓缩工艺研究

摘要 目的探索新疆紫草中以β,β′-二甲基丙烯酰阿卡宁为指标的制剂浓缩工艺的稳定性研究。方法采用动态平行取样,HPLC检测法,以β,β′-二甲基丙烯酰阿卡宁为考察指标,系统考察其热稳定性规律,用于浓缩过程中工艺参数的确定。结果当温度在60~80℃时,随加热温度升高,紫草提取液中阿卡宁的保留率降低;在固定温度下,随加热时间的延长,阿卡宁的保留率降低;当乙醇浓度为10%~80%时,在含醇量一定的条件下,随加热时间的延长,阿卡宁的保留率降低;当乙醇浓度低于30%时,阿卡宁在溶液中极不稳定。结论紫草药材提取后进行浓缩时,温度应控制在65℃以下,采用少量、短时间的方式进行浓缩。 OBJECTIVE To study the stability of standard in the concentration process ofβ,β′-dimethylacrylalkannin solution of alcohol in Lithospermum erythrorhizon.METHODS Using dynamic parallel sampling,HPLC method,andβ,β′-dimethylacrylalkannin as the detection indicators to study the thermal stability during the concentration process,so as to determin the process parameters in this step.RESULTS The stability experimental results showed when the temperature was 60-80℃,the retention rate ofβ,β′-dimethylacrylalkannin decreased with the increase of heating temperature;at a fixed temperature,with the extension of the heating time,the retention rate ofβ,β′-dimethylacrylalkannin decreased;when the ethanol concentration was 10%~80%,the retention rate ofβ,β′-dimethylacrylalkannin decreased with the extension of the heating time under the condition of a certain alcohol content;when the ethanol concentration was less than 30%,β,β′-dimethylacrylalkannin was extremely unstable in the solution.CONCLUSION During in the process of concentration,the concentration temperature of Lithospermum erythrorhizon extract should be controlled below 65℃,and concentrated in a small amount and concentration time should be controlled in shorter period.
