建立心益好胶囊中华蟾酥毒基、脂蟾毒配基的含量测定方法

Establish a Method for the Determination of Cinobufagin Venom Toad and Resibufogenin in XinYiHao Capsule

目的建立心益好胶囊中华蟾酥毒基、脂蟾毒配基含量的测定方法。方法高效液相色谱法,色谱柱为Agilent ZORBAX C_(18)(5μm,4.6 min×150 mm);流动相:0.5%磷酸二氢钾溶液-乙腈(50∶50)(用磷酸调pH值为3.2);紫外检测器,检测波长:296 nm;柱温:40℃;流速为0.8 mL·min^-1。结果华蟾酥毒基在5.27~105.4μg·mL^-1范围内与峰面积呈良好的线性关系(相关系数r=1);脂蟾毒配基在5.26~105.2μg·mL^-1范围内呈良好的线性关系(相关系数r=1)。华蟾酥毒基、脂蟾毒配基平均回收率分别为98.9%,99.0%;RSD分别为1.32%,1.03%,结论本方法精密度、稳定性和重复性良好,简便快速,分离良好,结果准确可靠,可作为心益好胶囊中华蟾酥毒基、脂蟾毒配基含量的测定方法。

OBJECTIVE To establish a method for the determination of Cinobufagin venom toad and Resibufogenin in XinYiHao Capsule.METHODS High performance liquid chromatography with reflux extraction of methanol.The chromatographic column was Agilent ZORBAX C 18(5μm,4.6 min×150 mm).Mobile phase:0.5%potassium dihydrogen phosphate solution-acetonitrile(50∶50)(Adjust pH with phosphoric acid to 3.2);UV detector,detection wavelength:296 nm;column temperature:40℃;flow rate was 0.8 mL·min-1.RESULTS In the range of 5.27-105.4μg·mL-1,the Cinobufagin venom toad showed a good linear relationship with the peak area(correlation coefficient r=1);the Resibufogenin showed a good linear relationship in the range of 5.26-105.2μg·mL-1.(Correlation coefficient r=1).The average recoveries of Cinobufagin venom toad and Resibufogenin were 98.9%and 99.0%respectively;RSD were 1.32%and 1.03%respectively.CONCLUSION This method has good precision,stability and repeatability,simple and fast,good separation.The results are accurate and reliable,which can be a method for the determination of Cinobufagin venom toad and Resibufogenin in XinYiHao Capsule.
