摘要
选用淡水型青海弧菌Q67为测试菌种,以96微孔板为样品测试载体,用高灵敏度的微孔板式发光检测仪进行检测,建立了一种全自动,高灵敏度,高通量的水质发光菌急性毒性测试方法。用该方法分别测试了汞,甲苯,三氯甲烷的水质发光菌急性毒性,用excel拟合了被测物质浓度(mg/L)与青海弧菌Q67相对发光强度(%)的线性方程,分别计算得出了汞,甲苯,三氯甲烷的半数效应浓度EC50值,利用96微孔板法进行水质发光菌急性毒性测试具有自动快速、高通量、数据准确等显著优势。
Using fresh water Vibrio qinghaiensis Q67 as the test strain and 96 micropore plate as the sample test carrier,a highly sensitive and high-throughput test method for acute toxicity of water luminescent bacteria was established.The acute toxicity of mercury,toluene and trichloromethane was tested by this method.The linear equation between the concentration of the tested substance(mg/L)and the relative luminous intensity(%)of Vibrio qinghaiensis Q67 was fitted by Excel.The EC50 values of half effective concentration of mercury,toluene and trichloromethane were calculated respectively.The acute toxicity test of luminescent bacteria in water by 96 micropore plate method was automatic and rapid,high throughput and accurate data.
