摘要
目的设计并制作一种多功能的高通量药物筛选芯片,开展复方木鸡颗粒及其各组分诱导HepG2细胞凋亡作用的研究,通过高通量筛选初步阐明复方木鸡颗粒抗肝癌的药效组分。方法运用该药物筛选芯片,以HepG2细胞形态、生长状态以及细胞活力等特征进行芯片的适用性考察;同时探讨复方木鸡颗粒及其各组分对HepG2细胞作用24、48和72 h后,对HepG2细胞凋亡影响。结果细胞在芯片上形态正常,生长状态良好,24、48、72 h后细胞存活率均在96%以上;经作用24、48和72 h后,复方木鸡颗粒、核桃楸皮黄酮、菟丝子黄酮、山豆根生物碱均有明显的诱导HepG2细胞凋亡的作用,与空白组相比,细胞凋亡坏死率差异均有统计学意义(P<0.05),且随着给药时间的延长,呈现出时-效关系。结论该芯片重复性及适用性良好,且样品和试剂消耗量较小,可实现高通量多通道平行检测,极大地简化了药效筛选实验的复杂性,为中药复方有效组分抗肿瘤作用的快速筛选提供一种较为便捷的方法。
Objective To design a multi-functional high-throughput drug screening chip to determine the apoptosis effect of Fufang Muji granules and its active components on HepG2 cells,and to preliminarily elucidate the pharmacodynamic components of Fufang Muji granules against liver cancer through high throughput screening.Methods The drug screening chip was used to investigate the applicability of the chip based on the morphology,growing status and cell viability of HepG2 cells.Meanwhile,the effects of Fufang Muji granules and their active components on the apoptosis of HepG2 cells after 24 h,48 h and 72 h were also determined.Results HepG2 cells on the chip grew in good condition,and the survival rate was above 96%after 24 h,48 h and 72 h.After 24 h,48 h and 72 h treament,Fufang Muji granules,flavonoids of Juglans mandshurica,lavonoids of Cuscuta chinensis and alkaloid of Sophora tonkinensis significantly induced the apoptosis of HepG2 cells.Compared with the blank group,the apoptotic and necrotic rates were significantly different(P<0.05),and showed a timeeffect relationship with the prolongation of treatment time.Conclusion The chip presents preferable repeatability and applicability,and the consumption of samples and reagents is small,which can realize high-throughput multi-channel parallel detection and simplify the complexity of efficacy screening experiments.This study is a convenient in the rapid screening of the anti-tumor effects of effective components of Chinese herbal compounds.
