摘要
目的评价柴胡不同极性部位的体外抗氧化能力,为柴胡的抗氧化作用研究及资源合理应用提供理论基础。方法制备柴胡的水、乙醇、乙酸乙酯、石油醚及正丁醇提取物,以维生素C为阳性对照,采用水杨酸法、DPPH法、ABTS法和普鲁士蓝法测定5种不同极性部位的体外抗氧化能力。结果柴胡5种极性部位均有较好的抗氧化能力,其中乙酸乙酯部位抗氧化活性最强,羟自由基、DPPH自由基、ABTS自由基的IC50分别为0.177、0.171、0.095 mg·mL-1,铁还原能力A0.5为0.491 mg·mL-1。各部位在不同体外抗氧化活性评价中的综合能力顺序为:乙酸乙酯部位>水部位>乙醇部位>正丁醇部位>石油醚部位。结论柴胡不同极性部位均具有一定的体外抗氧化能力,且抗氧化能力存在差异。
Objective To evaluate the antioxidant effect of different polar extracts of Bupleuri Radix in vitro and to provide a theoretical basis for the antioxidant activity of Bupleuri Radix and its rational use.Methods The in vitro antioxidant activities of the aqueous,ethanolic,ethyl acetate,petroleum ether,and n-butanol extracts of Bupleuri Radix were determined with salicylic acid,DPPH,ABTS,and Prussian blue assays.Vitamin C was used as the positive control.Results Different polar extracts of Bupleuri Radix showed good antioxidant activity,among which the ethyl acetate extract had the strongest activity.The IC50 values for the hydroxyl,DPPH,and ABTS radical scavenging activities were 0.177,0.171,and 0.095 mg·mL-1,respectively.The iron reduction capacity(A0.5)was 0.491 mg·mL-1.The potency of the extracts based on the in vitro antioxidant assays was as follows:ethyl acetate extract>water extract>ethanol extract>n-butanol extract>petroleum ether extract.Conclusion The different polar extracts of Bupleuri Radix exhibit different antioxidant effects in vitro.
