摘要 为缓解鹭鸶草(Diuranthera major)野生资源短缺状况,以鹭鸶草花序轴为外植体,研究不同消毒方式对外植体无菌培养的影响以及不同培养基配方对鹭鸶草组织培养的影响。结果表明,流水冲洗20 min、酒精消毒10 s、浓度1/1000氯化汞消毒8 min,花序轴污染率为15.33%,成活率为68.67%;MS+2.0 mg/L 6-BA+0.25 mg/L NAA可促进花序轴直接诱导出不定芽,诱导率为93.33%;适宜的增殖配方为MS+1.50 mg/L 6-BA+0.10 mg/L NAA+0.75 mg/L KT,增殖系数为6.8;在MS培养基中添加20 g/L NH4NO3和20 g/L土豆汁有助于壮苗,叶色翠绿且舒展度高;生根培养基为1/2 MS+0.50 mg/L IBA+0.20 mg/L NAA,转入该培养基中16~17天即可生根,平均生根数7.94条,生根率100%。以鹭鸶草花序轴为外植体可建立高效快繁再生体系,实现无性系规模化育苗,移栽成活率达100%。 In order to alleviate shortage of wild Diuranthera major resources,using rachis of D.major as ex⁃plant,effects of different sterilization methods on aseptic culture of explants and effects of different medium formulations on tissue culture of D.major were studied.Results showed that the contaminating rate of rachis was only 15.33%and survival rate was 68.67%after washing with running water for 20 min,soaking with al⁃cohol for 10 s and 1/1000 HgCl2 for 8 min to disinfection.Adventitious shoots could be directly induced from rachis in MS+2.0 mg/L 6-BA+0.25 mg/L NAA with induction rate 93.33%.The optimal proliferation medi⁃um was MS+1.50 mg/L 6-BA+0.10 mg/L NAA+0.75 mg/L KT,and the proliferation times was 6.8.Add⁃ing 20 g/L NH4NO3 and 20 g/L potato juice to MS was helpful to strengthen seedling,and leaves were more emerald green and spreading higher.Rooting medium was 1/2 MS+0.50 mg/L IBA+0.20 mg/L NAA,trans⁃ferring in which root could appear for 16-17 days,and the average rooting number was 7.94 with rooting rate 100%.Using rachis of D.major as explant could establish high efficiency and rapid propagation regeneration system,and realize large scale clonal seedling raising with survival rate of transplanting 100%.
