摘要
目的:探讨5-HT2A受体激动剂(DOI)对七氟醚(Sevo)或丙泊酚(PRO)麻醉联合Aβ_(1-40)诱导的大鼠术后认知功能障碍(POCD)的影响。方法:将50只SD大鼠随机分为5组:假手术组、Sevo麻醉联合Aβ_(1-40)诱导的POCD组(Sevo-POCD组)、PRO麻醉联合Aβ_(1-40)诱导的POCD组(PRO-POCD组)、DOI+Sevo-POCD组(在Sevo-POCD组处理基础上一次性静脉注射0.5 mg/kg的DOI)、DOI+PRO-POCD组(在PRO-POCD组处理基础上一次性静脉注射0.5 mg/kg的DOI)。采用水迷宫实验评估各组大鼠的认知功能,Garcia评分评价大鼠神经功能,苏木精-伊红(HE)染色法观察海马组织的病理学变化,TUNEL法检测神经元细胞凋亡,免疫荧光法检测5-HT2A受体的表达。结果:与假手术组比较,Sevo-POCD组和PRO-POCD组海马区神经组织损伤明显,神经元细胞凋亡率升高,逃避潜伏期延长,第Ⅱ象限停留时间缩短,穿越平台次数减少,Garcia评分降低,5-HT2A受体表达下调(均P<0.05);DOI+Sevo-POCD组、DOI+PRO-POCD组分别与Sevo-POCD组、PRO-POCD组比较,大鼠海马区神经组织损伤明显改善,神经元细胞凋亡率降低,逃避潜伏期缩短,第Ⅱ象限停留时间延长,穿越平台次数增加,Garcia评分升高,5-HT2A受体表达上调(均P<0.05)。结论:激活5-HT2A受体对Sevo和PRO麻醉联合Aβ_(1-40)诱导的大鼠POCD具有改善作用。
Objective:To investigate the effect of 5-HT2A receptor agonist(DOI)on postoperative cognitive dysfunction(POCD)induced by anesthesia sevoflurane(Sevo)or propofol(PRO)anesthesia combined with Aβ_(1-40).Methods:Forty 4-month-old SD rats were randomly divided into 5 groups:sham-operation group,Sevo-POCD group(POCD induced by Aβ_(1-40)under Sevo anesthesia),PRO-POCD group(POCD induced by Aβ_(1-40)under PRO anesthesia),DOI+Sevo-POCD group(One-time intravenous injection of 0.5 mg/kg DOI on the basis of Sevo-POCD group),DOI+PRO-POCD group(One-time intravenous injection of 0.5 mg/kg DOI on the basis of PROPOCD group).Water maze test was performed to evaluate cognitive function of rats.The neurological function of rats was evaluated by Garcia score.The pathological changes of hippocampal tissues were observed by hematoxylin-eosin(HE)staining.The neuronal apoptosis was detected by TUNEL assay.The expression of 5-HT2A receptor was detected by immunofluorescence assay.Results:Compared with the sham-operation group,the hippocampal nerve tissue was obviously damaged,the apoptosis rate of neurons was increased,the escape latency was prolonged,the second quadrant residence time was shortened,the times of crossing the platform was reduced,the Garcia score was decreased,and the expression of 5-HT2A receptor was down-regulated in Sevo-POCD and PRO-POCD groups(P<0.05).Compared with Sevo-POCD and PRO-POCD groups,the nerve tissue damage in the hippocampus of rats was significantly improved,the rate of neuronal apoptosis was decreased,the escape latency was shortened,the residence time in quadrant II was prolonged,the number of platform crossing was increased,Garcia score was increased,and the expression of 5-HT2A receptor was up-regulated after treated with DOI(P<0.05).Conclusion:Activation of HT2A receptors can ameliorate rat POCD induced by Sevo and PRO anesthesia combined with Aβ_(1-40).
