大蒜素对葡萄糖氯己定诱导的大鼠腹膜纤维化的影响 被引量:3

Effect of allicin on peritoneal fibrosis induced by glucose chlorhexidine in rats

摘要 目的探讨大蒜素对葡萄糖氯己定诱导的大鼠腹膜纤维化(PF)的影响。方法随机将30只SD大鼠分为空白组、模型组与大蒜素组,每组10只。模型组与大蒜素组大鼠以10 ml/kg的浓度腹腔注射0.1%葡萄糖氯己定溶液,空白组大鼠腹腔注射等量生理盐水,每天1次,持续21 d。大蒜素组大鼠于实验开始给予大蒜素溶液22 mg/kg灌胃,每天1次;空白组和模型组大鼠给予等量生理盐水灌胃,持续12周后处死所有大鼠。采用苏木素⁃伊红(HE)与马松(Masson)染色观察大鼠腹膜组织病理情况。采用免疫组化法检测大鼠腹膜组织中胶原蛋白Ⅲ(ColⅢ)、α⁃平滑肌肌动蛋白(α⁃SMA)、转化生长因子(TGF)⁃β1、Smad蛋白7(Smad7)、磷酸化细胞外调节蛋白激酶1/2(p⁃ERK1/2)的表达水平。采用实时荧光定量聚合酶链反应(RT⁃PCR)检测大鼠腹膜组织中微小RNA(miR)⁃21的表达水平。结果与模型组比较,大蒜素组大鼠腹膜组织增厚减轻,成纤维细胞、纤维细胞增生和炎性细胞浸润减少。与空白组比较,模型组大鼠纤维率明显提高,腹膜中ColⅢ、α⁃SMA、TGF⁃β1、Smad7、p⁃ERK1/2及miR⁃21表达水平明显升高(P<0.05);与模型组比较,大蒜素组大鼠纤维率明显降低,腹膜中ColⅢ、α⁃SMA、TGF⁃β1、Smad7及p⁃ERK1/2及miR⁃21表达水平明显降低(P<0.05)。结论大蒜素能减轻葡萄糖氯己定诱导的大鼠PF,miR⁃21的下调可能参与了大蒜素减轻PF的病理生理过程。 Objective To investigate the effect of allicin on peritoneal fibrosis(PF)induced by glucose chlorhexidine in rats.Methods Thirty SD rats were randomly divided into control group,model group and allicin group,with 10 rats in each group.Rats in the model group and allicin group were intraperitoneally injected with 0.1%glucose chlorhexidine solution at a concentration of 10 ml/kg,and rats in the control group were intraperitoneally injected with the same amount of normal saline once a day for 21 d.Rats in allicin group were given allicin solution 22 mg/kg intragastrically once a day after modeling,and rats in control group and model group were given the same amount of normal saline intragastrically.All rats were sacrificed after 12 weeks.The pathological condition of peritoneal tissue was observed by HE and Masson staining.The collagenⅢ(ColⅢ),α⁃SMA,TGF⁃β1,Smad7 and p⁃ERK1/2 expression levels were detected by immunohistochemistry in peritoneal.The miR⁃21 levels were detected by RT⁃PCR in peritoneal.Results Compared with the model group,peritoneal tissue thickening of allicin group was decreased,and fibroblast,fibroblast proliferation and inflammatory cell infiltration were decreased.Compared with the control group,PF rate in rats of model group increased significantly,the peritoneum ColⅢ,α⁃SMA,TGF⁃β1,Smad7,p⁃ERK1/2 and miR⁃21 expression level increased significantly(P<0.05).Compared with model group,the PF rate of allicin group was decreased,the levels of peritoneum ColⅢ,α⁃SMA,TGF⁃β1,Smad7,p⁃ERK1/2 and miR⁃21 in the allicin group were decreased significantly(P<0.05).Conclusion Allicin can alleviate glucose chlorhexidine induced PF in rats,and the down⁃regulation of miR⁃21 may be involved in the pathophysiological process of allicin alleviating PF.
