摘要
GRHL3是一种转录因子,为高度保守的Grainyhead家族成员之一。在胚胎发育过程中,其主要表达于外胚层;而在成年期,GRHL3也主要表达于外胚层来源的组织中,如皮肤和口腔等。GRHL3不仅在正常生长发育过程中发挥关键作用,如皮肤屏障形成、切口愈合、眼睑及神经管闭合、内皮细胞的迁移和凋亡等,而且参与了肿瘤相关靶基因和信号通路的调控,与肿瘤的增殖、凋亡、转移和侵袭等密切相关。然而,GRHL3在肿瘤中的作用机制十分复杂,且在人体不同类型恶性肿瘤的发生、发展过程中扮演不同角色,故对其深入研究有望为未来抗肿瘤治疗提供新方向。
Grainyhead-like 3(GRHL3),a transcription factor,is a member of the highly conservative Grainyhead family.During embryonic development,GRHL3 is mainly expressed in ectoderm,while in adult,GRHL3 is mainly expressed in ectoderm derived tissues,such as skin and oral cavity.GRHL3 not only plays important roles in the process of normal growth and development:such as the skin barrier formation,wound healing,eyelid and neural tube closure,endothelial cell migration and apoptosis,and so on,but also is involved in the regulation of tumor-related target genes and signaling pathways,and is closely related to the proliferation,apoptosis,metastasis and invasion of tumors.However,the mechanism of GRHL3 action in tumor is very complex,and it plays different roles in the occurrence and development of different types of malignant tumors,in-depth study of which is expected to provide a new direction for future anti-tumor therapy.
