基于Wilson病模型论肝主疏泄对铜稳态昼夜节律调控的现代研究构想

Modern Scientific Conception of Regulation of Copper Homeostasis Rhythm by Liver Controlling Conveyance and Dispersion based on Wilson’s Disease Model

铜稳态失调引发的功能障碍涉及多种疾病,铜维持生命不仅需要细胞中铜的稳态调节,而且需要铜转运到与其相应的配体中,避免氧化应激反应。线粒体也受到生物钟的控制,如果这些机制不能正常运行,将会导致铜的过剩或缺乏引起各种疾病。在肝主疏泄昼夜节律的调控下人体的五脏是应自然界四时阴阳消长而变化的时间自稳调节系统,在这一系列转化过程中,肝是五脏应时而变的内在推动力。因此,以Wilson病为疾病模型尝试从现代医学细胞生物学层面,结合时间医学相关研究结果与肝主疏泄功能和铜稳态平衡结合进行交叉研究以阐明肝主疏泄对线粒体铜稳态昼夜节律调控的科学内涵,对揭示铜稳态失调相关疾病机体整体调节规律的深层内涵具有重要意义。

The dysfunction caused by the dysregulation of copper homeostasis involves a variety of diseases.The maintenance of life by copper requires not only the homeostasis of copper in cells,but also the transport of copper to its corresponding ligands to avoid oxidative stress.Mitochondria are also controlled by the biological clock.If these mechanisms do not operate normally,it will lead to excess or deficiency of copper and cause various diseases.Under the regulation of the circadian rhythm of the liver governing the regulation of drainage,the five internal organs of the human body are a time self-regulating system that changes in response to the growth and decline of yin and yang at four o’clock in nature.In this series of transformation processes,the liver is the internal driving force for the five internal organs to change with time.Therefore,using Wilson’s disease as a disease model,an attempt was made to cross-study from the level of modern medical cell biology,combined with the results of time-related medical research,combined with the liver function and the balance of copper homeostasis,to clarify that the liver is responsible for the regulation of mitochondrial copper homeostasis.The scientific connotation of rhythm regulation is of great significance to reveal the deep connotation of the overall regulation of the body’s overall regulation of diseases related to copper homeostasis.
