益气化浊胶囊预防性给药对胰岛素抵抗大鼠肝脏的影响

Effect of Preventive Administration of Yiqi Huazhuo Capsule on the Liver of Rats with Insulin Resistance

目的:观察益气化浊胶囊预防性给药对胰岛素抵抗(IR)大鼠肝脏的影响。方法:SD大鼠随机分为空白对照组、模型对照组、盐酸二甲双胍组(0.2g/kg·d)和益气化浊胶囊高、中、低剂量组(2g/kg·d、1g/kg·d、0.5g/kg·d),每组10只。给药与造模同时进行,灌胃给药,每日1次,连续8周;造模方法采用高脂饮食诱导IR法。实验结束时,测量实验大鼠体重、肝脏湿重、计算肝脏指数,快速血糖仪测空腹血糖(FPG),生化法测定甘油三酯(TG)、胆固醇(TC)水平,酶联免疫吸附法(Elisa)检测血清胰岛素(FINS)水平,计算稳态模型胰岛素抵抗指数(HOMA-IR)评价IR水平。苏木精-伊红(HE)染色观察肝脏组织病理改变。结果:实验结束时,体重、肝脏湿重、肝脏指数、TG、TC、FINS、HOMA-IR模型对照组大鼠均显著高于空白对照组;体重盐酸二甲双胍组、益气化浊胶囊高剂量组大鼠均显著低于模型对照组大鼠,肝脏湿重、肝脏指数、TG、FINS、HOMA-IR盐酸二甲双胍组、益气化浊胶囊高剂量组、益气化浊胶囊中剂量组显著低于模型对照组大鼠,差异均有统计学意义。结论:益气化浊胶囊预防性给药在一定程度上可改善IR大鼠的肝脏脂肪病变,从而起到保护肝脏的作用。

Objective:To observe the effect of preventive administration of Yiqi Huazhuo Capsule on the liver of rats with insulin resistance(IR).Methods:SD rats were randomly divided into blank control group,model control group,metformin hydrochloride group(0.2g/kg·d)and Yiqi Huazhuo capsule high,medium and low dose groups(2g/kg·d,1g/kg)·D,(0.5g/kg·d),10 per group.The administration and modeling were carried out at the same time.Gavage was administered once a day for 8 consecutive weeks;the modeling method used high-fat diet to induce IR.At the end of the experiment,the experimental rats’body weight,liver wet weight,liver index were calculated,fasting blood glucose(FPG)measured by fast blood glucose meter,triglyceride(TG)and cholesterol(TC)levels determined by biochemical method,and enzyme-linked immunosorbent method(Elisa)detects serum insulin(FINS)levels,and calculates homeostatic model insulin resistance index(HOMA-IR)to evaluate IR levels.Hematoxylin-Eosin(HE)staining was used to observe the pathological changes of liver tissue.Results:At the end of the experiment,the body weight,liver wet weight,liver index,TG,TC,FINS,HOMA-IR model control rats were significantly higher than the blank control group;body weight metformin hydrochloride group,Yiqi Huazhuo capsule high-dose group The rats were significantly lower than the model control group.The liver wet weight,liver index,TG,FINS,HOMA-IR metformin hydrochloride group,Yiqi Huazhuo Capsule high-dose group,and Yiqi Huazhuo Capsule middle-dose group were significantly lower Model control rats,the differences are statistically significant.Conclusion:The preventive administration of Yiqi Huazhuo Capsule can improve the liver fatty pathology of IR rats to a certain extent,thereby protecting the liver.
