摘要
目的利用肿瘤数据库挖掘数据分析DCTPP1基因在乳腺癌组织中的表达及与患者预后的相关性,探讨DCTPP1基因的生物学作用及功能。方法利用ualcan数据库分析乳腺癌组织中的DCTPP1基因mRNA表达水平,利用Human Protein Reference Database数据库分析不同病理类型乳腺癌组织中DCTPP1蛋白表达水平,采用GEPIA乳腺癌数据分析DCTPP1基因的表达水平与乳腺癌患者预后的相关性。在genemania和WebGestalt数据库对与DCTPP1基因表达相关基因及其功能进行生物信息学分析,探讨基因功能。结果与正常乳腺组织比较,乳腺癌组织中DCTPP1基因mRNA呈高表达(P<0.05)。不同病理类型、不同肿瘤分期的乳腺癌组织中DCTPP1基因均呈高表达(P<0.05)。乳腺癌组织中DCTPP1蛋白表达水平高于正常乳腺组织。与低表达比较,DCTPP1基因高表达明显降低乳腺癌患者总体生存时间(HR=1.9,P<0.05)。基因功能分析提示与DCTPP1表达相关的20个基因主要位于细胞质、细胞核及细胞膜性结构中,参与细胞的代谢、生物调节、细胞生长等过程,在结合蛋白、酶活性、结合核酸等过程中发挥着作用。结论DCTPP1基因在乳腺癌中明显高表达,与乳腺癌患者不良预后相关,参与多种生物过程,可能成为乳腺癌治疗干预的新靶点。
Objective To analyze the expression of DCTPP1 in breast cancer and its correlation with the prognosis of breast cancer patients.To explore the biological function of DCTPP1.Methods The expression level of DCTPP1 mRNA in breast cancer tissues was analyzed by using the data base of ualcan.Human Protein Reference Database was used to analyze the expression of DCTPP1 in different pathological types of breast cancer.GEPIA breast cancer data were used to analyze the correlation between the expression level of DCTPP1 gene and the prognosis of breast cancer patients.The genes related to DCTPP1 expression and their functions were analyzed by bioinformatics in genemania and webgestalt databases.Results Compared with normal breast tissue,DCTPP1 mRNA was highly expressed in breast cancer(P<0.05).DCTPP1 was highly expressed in breast cancer tissues of different pathological types and tumor stages(P<0.05).The expression level of DCTPP1 protein in breast cancer was higher than that in normal breast tissue.Compared with the low expression patients,the high expression of DCTPP1 significantly reduced the overall survival time of breast cancer patients(HR=1.9,P<0.05).The analysis of gene function indicated that the 20 genes related to DCTPP1 expression were mainly located in the cytoplasm,nucleus and cell membrane structure,which were involved in cell metabolism,biological regulation,cell growth and other processes.It plays an important role in binding protein,enzyme activity and nucleic acid.Conclusion DCTPP1 is highly expressed in breast cancer,which is related to the poor prognosis of breast cancer patients.DCTPP1 may be a new target of breast cancer treatment and intervention.
