摘要
基于三维荧光光谱仪以及斜投影算法,建立了养殖水体中孔雀石绿含量的快速分析模型。首先向样品中加入足量硼氢化钾粉末,将水溶性的孔雀石绿(MG)完全转化为不溶于水的隐性孔雀石绿(LMG),然后加入等体积的乙腈溶解LMG,通过三维荧光检测LMG的含量进而转化得到养殖水体中MG的含量,可提高检测灵敏度。结果表明:基于斜投影算法进行化学计量学分析,可以减少样品的假阴性率和假阳性率,测定结果与高效液相色谱法测定结果一致,与平行因子算法相比较,斜投影算法的相对误差较小,LMG标准品在0.02~0.60μg·mL^(-1)浓度范围内线性关系良好,相关系数r=0.9984,检出限为1.28×10^(-2)μg·mL^(-1),加标回收率在83.33%~100.00%之间,精密度RSD≤0.55%。三维荧光光谱仪检测灵敏度高,样品采集快速,结果准确可靠,可用于多组分混合物的检测分析。
A method for the rapid detection and analysis of malachite green in aguiculture water was established based on the three-dimensional fluorescence spectrometer and oblique projection algorithm.Firstly,eddingenough potassium borohydride powder to the samples can convert the water-soluble malachite green(MG)into the insoluble malachite green(LMG)completely.Secondly,equal volume of acetonitrile were added in order to dissolve the LMG in water.Finally,the content of LMG was detected by three-dimensional fluorescence and then converted to the content of MG in the aquaculture water,which can improve the detection sensitivity.The results showed that chemometric analysis combined with the oblique projection algorithm can reduce the false negative and false positive rates of the samples.The results of oblique projection algorithm were more silimilar to that of HPLC and had smaller relative errors compared with the parallel factor algorithm.A good linear relationship of LMG existedat the range of 0.02~0.60μg·mL^(-1),with the correlation coefficient r=0.9984,the detection limit was 1.28×10^(-2)μg·mL^(-1),the recovery rates were 83.33%~100.00%,and the precision RSD≤0.55%.The three-dimensional fluorescence spectrometer has high detection sensitivity,rapid sample collection,accurate and reliable results,and can be used for the detection and analysis of multi-component mixtures.
