摘要
目的检测在高糖培养条件下人视网膜血管内皮细胞中hsacirc0005699的表达,为糖尿病性视网膜病变发病机制研究提供新的线索。方法人视网膜血管内皮细胞分为低糖组(L组)、高渗对照组(C组)和高糖组(H组),设置低糖组中糖浓度为5.5mmol/L,高渗对照组中葡萄糖浓度为5.5mmol/L,甘露醇浓度为19.5mmol/L,高糖组中葡萄糖浓度为25mmol/L,三组均培养24hr、72hr、96hr。通过实时定量PCR(q-PCR)对差异表达的hsacirc0005699进行验证。最后采用t-检验、相关分析等方法分析实验结果。结果q-PCR结果表明,hsacirc0005699在24hr、72hr和96hr高糖培养的视网膜血管内皮细胞中表达都显著升高(P<0.05),在高渗对照组中表达无统计学意义。结论hsacirc0005699在高糖培养的人视网膜血管内皮细胞中表达显著升高,有望成为糖尿病性视网膜病变潜在研究方向。
Objective To detect the expression of hsacirc0005699 in human retinal vascular endothelial cells cultured with high glucose,and provides a new clue for the study of the pathogenesis of diabetic retinopathy.Methods Human retinal vascular endothelial cells are divided into low glucose group(L group),hypertonic control group(C group)and high glucose group(H group).Set the glucose concentration of the low group was 5.5 mmol/L,the glucose concentration of the hypertonic control group was 5.5 mmol/L,the mannitol concentration was 19.5 mmol/L,the glucose concentration of the high glucose group was 25 mmol/L,and all of the three groups were cultured for 24 hr and 72 hr、96 hr.The differentially expressed of hsacirc0005699 was verified by realtime quantitative PCR(q-PCR).Finally,t-test,correlation analysis and other methods are used to analyze the experimental results.Results The real-time fluorescent quantitative PCR results showed that the expression of hsacirc0005699 in the retinal vascular endothelial cells cultured with high glucose was significantly increased for 24 hr,72 hr and 96 hr(P<0.05),and the expression in the hypertonic control group was not statistically significant.Conclusions The expression of hsacirc0005699 is significantly increased in human retinal vascular endothelial cells cultured with high glucose,and it is expected to become a potential research direction for diabetic retinopathy.
