含CC31毕赤酵母工程菌固态发酵生产蛋白质饲料的效果 被引量:1

摘要 为研究含抗菌肽CC31毕赤酵母工程菌固态发酵生产高功能性蛋白质饲料工艺的优化,采用单因素试验设计筛选豆粕和麦麸最佳粒度;在最佳粒度的前提下利用DPS软件中均匀设计模型,拟合小分子蛋白浓度(Y)与接种量(X_(1))、含水量(X_(2))相关曲线。发酵结束测定小分子蛋白浓度并与预测值进行比较。同时对发酵饲料的营养成分进行分析。结果表明:最佳发酵粒度为40~60目,小分子蛋白浓度拟合曲线:Y=113.8745+0.9155 X_(1)+1.1952 X_(2)-0.0465 X_(1)·X_(1)-0.1167 X_(2)·X_(2),R^(2)=0.99,通过拟合曲线预测得出当接种量X_(1)为9.85%,含水量X_(2)为51.22%,小分子蛋白浓度(Y)的含量最高为148.99 mg·L^(-1)。依据拟合曲线的最优接种量和含水量,设置接种量X_(1)为10%,含水量X_(2)为50%,试验得出小分子蛋白浓度为156.23 mg·L^(-1)。与模拟曲线预测比较相对误差仅为4.63%。对其常规营养成分分析,发现能有效的增加粗蛋白含量,但对纤维的分解效果不明显。 To study the optimization of the process of solid-state fermentation of high-performance protein feed by Pichia pastoris engineered bacteria containing the antibacterial peptide CC31,single factor test design was used to screen the optimal particle size of soybean meal and wheat bran.Based on the optimal particle size,the model was uniformly designed in DPS software to fit the correlat-ion curves of small molecule protein concentration(Y)with inoculation amount(X_(1))and water content(X_(2)).The concentration of small molecule protein was determined at the end of the fermentation and compared with the predicted value.At the same time,the nutritional composition of the fermented feed was analyzed.The results showed that the optimal fermentation particle size was 40 to 60 meshes,and the small molecule protein concentration fitting curve:Y=113.8745+0.9155 X_(1)+1.1952 X_(2)-0.0465 X_(1)·X_(1)-0.1167 X_(2)·X_(2),R^(2)=0.99.It was predicted by the fitted curve that when the inoculation amount X_(1) is 9.85%,the water content X_(2) was 51.22%,and the content of the small molecule protein concentration(Y)was 148.99 mg·L^(-1).According to the optimal inoculation amount and water content of the fitted curve,the inoculation amount X_(1) was set to 10%,and the water content X_(2) was set to 50%.The test obtained a small molecule protein concentration of 156.23 mg·L^(-1).Compared with the simulation curve prediction,the relative error was only 4.63%.By testing the conventional nutritional components of the substrate after fermentation,it was found that the crude protein content could be effectively increased,but the effect on fiber decomposition was not obvious.
