摘要
目的通过检测非小细胞肺癌(NSCLC)组织中长链非编码RNA(LncRNA)CRNDE的表达水平,探讨其对NSCLC细胞增殖、迁移及侵袭的影响及其分子机制。方法选择2018年1月至2019年6月在孝昌县第一人民医院胸外科收治的30例行手术切除的NSCLC患者,其中男性17例,女性13例;年龄34~75岁,中位年龄58岁;病理组织学类型,腺癌12例,鳞癌15例,鳞腺癌3例。收集入组患者癌组织及对应癌旁正常组织(距癌组织>5 cm)标本,利用荧光定量聚合酶链反应(RT-qPCR)检测结直肠癌差异表达基因(CRNDE)在NSCLC组织中的表达。NSCLC细胞PC-9中转染pcDNA-CRNDE和对照pcDNA,RT-qPCR验证其转染效率。利用CCK-8法、Transwell实验研究CRNDE过表达对PC-9细胞增殖、侵袭及迁移的影响。Western blot检测PI3Kp85α(磷脂酰肌醇3-激酶p85α亚单位)、p-Akt及T-Akt蛋白表达水平。结果NSCLC组织中LncRNA CRNDE的相对表达水平(1.643±0.425)显著高于癌旁正常组织(0.967±0.286)(t=7.228,P<0.001)。NSCLC细胞系中LncRNA CRNDE表达显著升高(P<0.05)。转染pcDNA-CRNDE后细胞中CRNDE表达水平显著高于转染pcDNA细胞(P<0.001)。过表达CRNDE显著促进了NSCLC细胞PC-9的增殖、迁移及侵袭能力(P<0.05)。过表达CRNDE显著上调了NSCLC细胞PC-9中p-Akt/T-Akt和PI3Kp85α蛋白表达水平(P<0.01)。结论NSCLC组织中CRNDE显著高表达,过表达CRNDE可促进NSCLC细胞的生物学行为,可能通过激活PI3K/Akt信号通路来实现。
Objective To detect expression level of long non-coding RNA(LncRNA)colorectal neoplasia differentially expressed(CRNDE)in non-small cell lung cancer(NSCLC)tissues,and explore its effects on proliferation,migration and invasion of NSCLC cells and their molecular mechanisms.Methods A total of 30 cases of NSCLC patients who underwent surgical resection from January 2018 to June 2019 were enrolled,which included 17 males and 13 females,aged 34-75 years old with medium age of 58 years old;histological types were 12 cases with adenocarcinoma,15 with squamous cell carcinoma and 3 with squamous adenocarcinoma.The cancer tissue and adjacent normal tissue(>5 cm from cancer tissue)specimens were collected,and fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect expression of CRNDE in NSCLC tissues.NSCLC cells PC-9 were transfected with pcDNA-CRNDE and control pcDNA,and transfection efficiency was verified by RT-qPCR.CCK-8 method and Transwell experiment were used to study the effect of CRNDE overexpression on PC-9 cell proliferation,invasion and migration.Protein expression levels of PI3Kp85α(phosphatidylinositol 3-kinase p85αsubunit),p-Akt and T-Akt were detected by Western blot.Results The relative expression level of LncRNA CRNDE in NSCLC tissue(1.643±0.425)was significantly higher than that in adjacent normal tissues(0.967±0.286)(t=7.228,P<0.001).LncRNA CRNDE expression was significantly increased in NSCLC cell lines(P<0.05).The expression level of CRNDE in pcDNA-CRNDE transfected cells was significantly higher than that in pcDNA-CRNDE transfected cells(P<0.001).The overexpression of CRNDE significantly promoted proliferation,migration and invasion of PC-9 in NSCLC cells(P<0.05).The overexpression of CRNDE significantly increased expression levels of p-Akt/T-Akt and PI3Kp85αin PC-9 cells of NSCLC(P<0.01).Conclusion It is demonstrated that CRNDE is significantly highly expressed in NSCLC tissues,and overexpression of CRNDE can promote biological behavior of NSCLC cells,which may be achieved by activating PI3K/Akt signaling pathway.
