摘要
目的:基于网络药理学的方法,探讨海昆肾喜胶囊治疗慢性肾衰竭的潜在作用机制。方法:查找海昆肾喜胶囊有效成分,获取褐藻多糖硫酸酯(Fucoidan)的2D和3D结构并预测药效团的靶点,加上基因名称后构建蛋白互作PPI网络,绘制出"有效成分-靶点基因-通路"的网络图,进行GO功能富集分析和KEGG通路富集分析。结果:筛选得到褐藻多糖硫酸酯有基因名称的靶点287个,可作用于300个潜在靶点和113条相关通路,涵盖调节内分泌、抗疾病和代谢通路等多个环节,而这些功能和通路与肾脏疾病密切相关。结论:通过网络药理学药效团分析,推测海昆肾喜胶囊是通过多靶点、多通路作用于人体,从而发挥治疗CRF的作用。
Objective:To investigate the potential mechanism of action of Haikun Shenxi capsules in the treatment of chronic renal failure based on network pharmacology.Methods:The effective constituents of Haikun Shenxi capsules were obtained.The 2D and 3D structures of fucoidan were obtained,and the targets of pharmacophore were predicted.A protein-protein interaction network was established after the names of genes were added,and an"effective constituent-target gene-pathway"network was plotted.Gene ontology(GO)enrichment analysis and KEGG pathway enrichment analysis were performed.Results:A total of 287 targets of fucoidan with gene names were screened out,which acted on 300 potential targets and 113 related pathways,covering various links in endocrine regulation,disease prevention,and metabolism pathways,and these functions and pathways were closely associated with renal diseases.Conclusion:Based on network pharmacology and pharmacophore,it is predicted that Haikun Shenxi capsules exert a therapeutic effect on chronic renal failure by acting on multiple targets and pathways.
