芝麻素对冠心病大鼠心肌细胞凋亡的影响及PKCα/NF-κB信号途径的作用研究 被引量:5

摘要 目的分析芝麻素对冠状动脉粥样硬化性心脏病(冠心病)大鼠心肌细胞凋亡及PKCα/NF-κB信号途径的影响。方法选取30只SPF级雄性SD大鼠分为对照组、模型组和观察组,每组各10只。模型组和观察组大鼠予以高脂饲料连续饲养16周,对照组给予基础饲料喂养。观察组造模完成后芝麻素灌胃治疗1/d,对照组和模型组使用等体积生理盐水进行干预。干预结束后禁食12 h检测三酰甘油(TG)、总胆固醇(TC)和高密度脂蛋白胆固醇(HDL-C)水平。以乙酰胆碱(ACH)和酸化亚硝酸钠刺激大鼠主动脉并记录血管张力变化。使用TUNEL法检测心肌细胞凋亡情况。采用Westernblot法检测心肌组织中Caspase-3。结果观察组大鼠血中TC水平明显低于模型组,差异具有统计学意义(P<0.05);观察组大鼠经ACH刺激后血管最大舒张程度明显低于模型组,差异具有统计学意义(P<0.05);观察组心肌细胞凋亡率明显低于模型组,Caspase-3和Caspase-12蛋白表达水平明显低于模型组,且差异具有统计学意义(P<0.05);观察组心肌细胞p-PKCα、p-IκBα和NF-κB p65蛋白表达水平明显低于模型组,差异具有统计学意义(P<0.05)。结论芝麻素通过调控PKCα/NF-κB信号途径有效抑制冠心病大鼠的心肌细胞凋亡。 Objective To analyze the influence of sesamin on cardiomyocyte apoptosis and PKCα/NF-κB signaling pathway in rats with coronary heart disease(CHD).Methods Male SD rats(SPF grade,n=30)were divided into control group,model group and observation group(each n=10).The model group and observation group were raised with high-fat diet and control group was raised with basic diet for 16 weeks.After modeling,the observation group was intragastrically given sesamin once a day,and control group and model group was given isometric normal saline for 6 weeks.After intervention finished and fasting for 12 h,the levels of TG,TC and HDL-C were detected.The changes of angiotasis were recorded through stimulating rat aorta with acetylcholine(ACH)and acidic nitrite.The cardiomyocyte apoptosis was detected by using TUNEL method,and level of myocardial Caspase-3 was detected by using Western blotting assay.Results The level of TC was significantly lower in observation group than that in model group(P<0.05).The maximum vasodilatation degree after ACH stimulation was significantly lower in observation group than that in model group(P<0.05).The cardiomyocyte apoptosis rate was significantly lower,and protein expressions of Caspase-3 and Caspase-12 were significantly lower in observation group than those in model group(P<0.05).The protein expressions of cardiomyocyte p-PKCα,p-IκBαand NF-κB p65 were significantly lower in observation group than those in model group(P<0.05).Conclusion Sesamin can effectively inhibit cardiomyocyte apoptosis through contolling PKCα/NF-κB signaling pathway in CHD rats.
