摘要
本实验首先对嗜酸乳杆菌进行相应的处理,使其复苏和活化,在进行生长曲线测定的过程中,发现其生长28h左右达到稳定期。之后将以28h为培养时间的L.a.s按不同体积分数作用于PC12细胞,荧光染色和CCK-8细胞增殖检测结果表明,PC12细胞能够正常生长,研究发现加入L.a.s体积分数为6%时,PC12细胞的存活率提高至对照组的131.31±4.59%。说明适宜浓度下的L.a.s具有促进PC12细胞生长的作用,为后续进一步研究其神经保护作用打下基础。
In this experiment,Lactobacillus acidophilus was treated to make it resuscitate and activate.In the process of measuring the growth curve,it was found that the growth of Lactobacillus acidophilus reached a stable stage in about 28 hours.After that,the PC12 cells were treated with L.a.s for 28h at different volume fractions.Fluorescence staining and CCK-8 cell proliferation test showed that PC12 cells could grow normally.It was found that the survival rate of PC12 cells increased to 131.31±4.59% of the control group when the volume fraction of L.a.s was 6%.The results indicated that L.a.s could promote the growth of PC12 cells at suitable concentration,which laid a foundation for further study of its neuroprotective effect.
