北京芽孢杆菌和节杆菌混合培养条件优化研究

摘要 试验旨在对北京芽孢杆菌和节杆菌混合培养条件进行单因素与响应面法优化。通过单因素试验分别确定接种量(2%、3%、4%)、温度(32℃、37℃、42℃)、初始pH值(6.2、7.4、8.6)及摇床转速(130 r/min、170 r/min、210 r/min)对北京芽孢杆菌和节杆菌(1∶1)混合培养的影响,应用响应面法对混菌培养条件进行优化。最佳优化条件为:接种量2%、pH 7.4、温度35.1℃、摇床转速210 r/min,此时可获得最大的预测吸光度值0.721,与优化前的吸光度值(0.442)相比提高63.35%,表明该模型能有效预测混菌发酵增殖培养情况。 This experiment was designed to optimize the mixed cultivation conditions of Bacillus beijingensis and Arthrobacter sp.by single factor and response surface methodology.In this study,the effects of inoculation amount(2%,3%,4%),temperature(32℃,37℃,42℃),initial pH(6.2,7.4,8.6)and shaker speed(130 r/min,170 r/min,210 r/min)on the mixed cultivation of Bacillus beijingensis and Arthrobacter sp.(1∶1)were determined by single factor test,and the cultivation conditions were optimized by Box-Behnken design of response surface methodology.The optimal optimization conditions were as follows:inoculation amount 2%,initial pH 7.4,temperature 35.1℃ and shaker speed 210 r/min.Under these conditions,the maximum predicted absorbance value was 0.721,which was 63.35%higher than the absorbance value before optimization(0.442),indicating that the model could effectively predict the proliferation and cultivation of mixed bacteria fermentation.
