摘要
借助生物信息学技术预测分析卷枝毛霉苹果酸转运(malate transporter,MT)蛋白的理化特性、空间结构及功能位点,通过Protpara、PredictProtein、TMHMM Server、NetPhos等在线分析工具对MT蛋白的基本性质和结构进行分析预测。结果表明,MT蛋白疏水性强且性质稳定,属于非分泌型蛋白,由10个跨膜螺旋逆时针排列形成一个接近于椭球形的蛋白质分子三维结构;该转运蛋白有8个豆蔻酰化位点,3个ASN-糖基化位点和蛋白激酶C磷酸化位点,1个酪蛋白激酶II磷酸化位点;系统发育分析发现,卷枝毛霉MT蛋白与毛霉菌(Mucor ambiguous)的同源性最高,聚为一类,与植物拟南芥(Arabidopsis thaliana)的亲缘关系较远。本研究为后续探究苹果酸转运蛋白的三维结构与功能奠定了基础。
In this study,the physical and chemical properties,spatial structure and functional sites of malate transporter(MT)protein from Mucor circinelloides were predicted and analyzed by means of bioinformatics technology,and the basic properties and structures of MT protein were predicted by using online analysis tools such as Protpara,PredictProtein,TMHMM Server and NetPhos.The results showed that MT protein was hydrophobic and stable,and belonged to non-secretory protein.The three-dimensional structure of protein molecule was similar with an ellipsoid formed by counterclockwise alignment of 10 transmembrane helices.There are eight N-myristoylation site sites,three ASN-glycosylation sites and protein kinase C phosphorylation sites,and one casein kinase II phosphorylation site.Phylogenetic analysis revealed that MT protein in M.circinelloides had the highest homology with Mucor ambiguus,but distantly related to Arabidopsis thaliana.This study may lay a foundation for the subsequent exploration of the three-dimensional structure and function of malate transporter.
