支气管哮喘患儿急性期血清miR-218、CCL26表达及临床意义 被引量:8

Expressions and clinical significance of miR-218 and CCL26 in serum of children with asthma at acute stage

摘要 目的检测支气管哮喘患儿血清中miR-218、CCL26表达水平,探究二者在支气管哮喘急性期表达的临床意义。方法选取支气管哮喘急性期患儿63例作为观察组,根据病情严重程度的患儿分为:轻度组25例,中度组20例,重度组18例;另选取入院复查的哮喘缓解期患儿22例作为对照组。采用qRT-PCR法检测两组受试者血清中miR-218表达水平;采用酶联免疫吸附(ELISA)法检测患儿血清中CCL26及IL-13、IL-17、IgE表达水平;分析不同严重程度患儿血清中miR-218、CCL26表达水平;Pearson法分析支气管哮喘患儿血清中miR-218、CCL26表达及与相关炎性因子的相关性。结果观察组患儿血清miR-218、FEV1、PEF水平显著低于对照组(P<0.05),CCL26、IL-13、IL-17、IgE水平显著高于对照组(P<0.05);中度组、重度组患儿血清miR-218水平显著低于轻度组(P<0.05),CCL26水平显著高于轻度组(P<0.05),重度组患儿血清miR-218水平显著低于中度组(P<0.05),CCL26水平显著高于中度组(P<0.05);支气管哮喘患儿血清miR-218水平与CCL26、IL-13、IL-17、IgE呈负相关(r=-0.614、-0.610、-0.678、-0.498,P<0.05),与FEV1、PEF呈正相关(r=0.680、0.643,P<0.05);CCL26与FEV1、PEF呈负相关(r=-0.659、-0.714,P<0.05),与IL-13、IL-17、IgE呈正相关(r=0.529、0.635、0.522,P<0.05)。结论支气管哮喘急性期患儿血清中miR-218呈低表达,CCL26呈高表达,二者与患者严重程度及炎性反应密切相关。 Objective To detect the expression levels of miR-218 and CCL26 in the serum of children with asthma and to explore the clinical significance of their expressions at acute stage of asthma.Methods From June 2018 to June 2019,85 children with asthma enrolled in our hospital were selected.Among them,63 asthma children at acute stage were in the acute stage group and 22 asthma children at remission stage were in the remission stage group.According to the severity of the disease,the children were divided into:25 cases in the mild group,20 cases in the moderate group and 18 cases in the severe group.The expression of miR-218 in serum of the two groups was detected by qRT-PCR.The expression levels of CCL26,IL-13,IL-17 and IgE in serum were detected by enzyme-linked immunosorbent assay(ELISA).The expression levels of miR-218 and CCL26 in serum of children with different severity were analyzed,and the expressions of miR-218 and CCL26 in the serum of asthmatic children and their correlations with the related inflammatory factors were analyzed by Pearson′s method.Results The levels of serum miR-218,FEV 1 and PEF in the observation group were significantly lower than those in the control group(P<0.05),and the levels of CCL26,IL-13,IL-17 and IgE were significantly higher than those in the control group(P<0.05).The level of serum miR-218 in the moderate group and the severe group was significantly lower than that in the mild group(P<0.05),and the level of CCL26 was significantly higher than that in mild group(P<0.05).The level of serum miR-218 in the severe group was significantly lower than that in the moderate group(P<0.05),and the level of CCL26 was significantly higher than that of the moderate group(P<0.05).The levels of miR-218 were negatively correlated with CCL26,IL-13,IL-17 and IgE(r=-0.614,-0.610,-0.678,-0.498,P<0.05),and positively correlated with FEV 1 and PEF(r=0.680,0.643,P<0.05).CCL26 was negatively correlated with FEV 1 and PEF(r=-0.659,-0.714,P<0.05),and positively correlated with IL-13,IL-17 and IgE(r=0.529,0.635,0.522,P<0.05).Conclusion The expression of miR-218 in serum of children with asthma at acute stage is low,while the expression of CCL26 is high,which is closely related to the severity and inflammatory response of the patients.
