参芍养胃膏制备工艺优选及初步药效学研究

Study on the optimization of extraction technology and preliminary pharmacodynamics of Shenshao Yangwei Decoction

目的优选参芍养胃膏的制备工艺条件,并初步评价其对功能性消化不良大鼠胃肠动力的影响,为该制剂的开发提供依据。方法采用正交试验,以芍药苷质量浓度及固形物质量为综合评分指标,考察提取时间、提取次数及加水倍量对参芍养胃膏提取工艺的影响;以芍药苷保留率为指标,考察浓缩终点;并通过收膏工艺考察确定赋形剂用量。通过观察参芍养胃膏对功能性消化不良大鼠胃排空率、小肠推动率的影响,从而探讨参芍养胃膏的治疗作用。结果最佳提取工艺为:水提取2次,每次提取1 h,加水量为6倍;减压浓缩至相对密度约为1.15(60℃)的稠膏,再加入阿胶、麦芽糖,制成相对密度为1.33~1.37(60℃)的膏滋。与模型组比较,参芍养胃膏组大鼠胃排空率及小肠推进率均明显提高(P<0.05)。结论优选的工艺稳定可行,药效学研究结果提示参芍养胃膏对功能性消化不良有一定的治疗作用。

Objective To optimize the preparation conditions of Shenshao Yangwei decoction,and preliminarily evaluate its effect on gastrointestinal motility of rats with functional dyspepsia,so as to provide basis for the development of the preparation.Methods Taking the content of paeoniflorin and the quality of solid substance as the comprehensive evaluation index,the effects of extraction time,extraction times and water times on the extraction process of Shenshao Yangwei decoction were investigated by orthogonal test.The concentration end point was determined by the retention rate of paeoniflorin,and the amount of excipient was determined by the technology of collecting ointment.The therapeutic effect of Shenshao Yangwei decoction was explored by observing the effect of Shenshao Yangwei decoction on gastric emptying rate and small intestine promoting rate of rats with functional dyspepsia.Results The best water extraction process was as follows:6 times for twice,each time 1 h.The solution was concentrated to a thick paste with a relative density of about 1.15(60℃)under reduced pressure,after adding donkey hide gelatin and maltose,the paste was concentrated to a relative density of 1.33-1.37(60℃).Compared with the model group,the gastric emptying rate and small intestine propulsion rate in Shenshao Yangwei decoction group were significantly increased.Conclusion The optimized technology is stable and feasible,and the results of pharmacodynamic study suggest that Shenshao Yangwei decoction has a certain therapeutic effect on functional dyspepsia.
