离子色谱法测定糊精中的5种还原糖 被引量:1

Determination of five reducing sugars in dextrin by ion chromatography

摘要 目的建立离子色谱法分离测定糊精中的还原糖。方法采用Dionex CarboPac PA200柱;以纯化水为流动相A,0.25 mol/L NaOH溶液为流动相B,0.25 mol/L醋酸钠溶液为流动相C,梯度洗脱;流速为0.4 mL/min,安培检测器,测定糊精中还原糖(以D‐葡萄糖,麦芽糖,麦芽三糖,麦芽四糖和麦芽五糖为代表)。结果糊精中D‐葡萄糖、麦芽糖、麦芽三糖、麦芽四糖和麦芽五糖分离度良好,在0.1~20μg/mL范围内线性关系良好,相关系数r分别为0.9999、0.9995、0.9992、0.9998、0.9996(n=6);定量限分别为7.5、7.9、11.8、12.3、7.5 ng/mL;平均回收率分别为98.4%、99.2%、100.9%、98.9%、99.1%。结论方法专属性强、准确、灵敏,可用于糊精中还原糖的测定。 Objective To establish an ion chromatography method for determination of reducing sugars in dextrin.Methods Dionex CarboPac PA200 was employed as anion analytical column.The chromatographic conditions were as follows:water as mobile phase A,0.25 mol/L sodium hydroxide solution as mobile phase B and 0.25 mol/L sodium acetate solution as mobile phase C with gradient elution at the flow rate of 0.4 mL/min.Amperometric detector was used to determine the content of reducing sugar(D‐glucose,maltose,maltotriose,maltotetraose and maltopentaose)in dextrin.Results D‐glucose,maltose,maltotriose,maltotetraose and maltopentaose were well separated.The results had good liner relationship within the range of 0.1-20μg/mL(r=0.9999,0.9995,0.9992,0.9998,0.9996 respectively,n=6).The quantitation limit of D‐glucose,maltose,maltotriose,maltotetraose and maltopentaose were 7.5、7.9、11.8、12.3 and 7.5 ng/mL(S/N=10),respectively.The average recoveries of D‑glucose,maltose,maltotriose,maltotetraose and maltopentaose were 98.4%、99.2%、100.9%、98.9%and 99.1%,respectively.Conclusion The method is specific,sensitive,accurate,which can be applied to determine the content of reducing sugar in dextrin.
