摘要
目的采用一测多评法测定白鲜皮药材中黄柏酮、白鲜碱、岑酮的含量,比较不同商品来源白鲜皮的质量。方法采用HPLC法,以黄柏酮为内参物建立其与白鲜碱、岑酮的相对校正因子,采用一测多评法和标准曲线法同时计算3种成分含量并进行比较;采用上述方法测定50批白鲜皮药材中3种成分的含量,并对结果进行聚类分析。结果白鲜皮药材中黄柏酮、白鲜碱、岑酮的含量分别为0.21%~0.53%、0.018%~0.061%、0.071%~0.171%,一测多评法的测定结果与标准曲线法无明显差异,RSD <2.0%。聚类分析法将50批白鲜皮药材分为3类,不同地区商品白鲜皮中3种成分的含量差异显著。结论所用方法操作简单,重复性好,可为白鲜皮的质量标准修订提供参考;聚类分析法对市场上白鲜皮的质量起到良好的评价作用,可为系统研究白鲜皮的品质奠定基础。
OBJECTIVE To establish a method for the determination of obacunone,dictamnine and fraxinellone in Dictamni Cortex,and to compare the quality of Dictamni Cortex from different commercial sources. METHODS The relative correction factors of obacunone with dictamnine and fraxinellone were established by HPLC,and the contents of the three components were calculated and compared using the method of multiple-evaluation and standard curve. Combined with the above methods,50 batches of Dictamni Cortex were determined,and the results were analyzed by the cluster analysis. RESULTS The contents of obacunone dictamnine and fraxinellone in Dictamni Cortex were 0. 2-0. 53%,0. 018-0. 061% and 0. 07-10. 171%,respectively. There was no significant difference( RSD < 2. 0%) between the multi-determination method and standard curve method. By cluster analysis,50 batches of Dictamni Cortex from different producing areas were divided into 3 categories,and the contents of Dictamni Cortex from different areas were significantly different. CONCLUSION The multi-evaluation method is simple and repeatable,which can provide reference for revising the quality standard of Dictamni Cortex,and the cluster analysis method plays a good role in evaluating the quality condition of Dictamni Cortex on the market. It can lay a foundation for further systematic study of Dictamni Cortex.
