摘要
目的比较不同采收期及生长年限独活中蛇床子素和二氢欧山芹醇当归酸酯的含量。方法采用HPLC,色谱柱为Agilent C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(49∶51),检测波长330 nm,流速1.0 mL·min^(-1),柱温30℃。结果 2~6年生春初采收的独活中蛇床子素和二氢欧山芹醇当归酸酯的含量分别为0.39%~1.11%、0.042%~0.185%;2~6年生秋末采收的独活中的含量则分别为0.34%~0.98%、0.034%~0.177%。蛇床子素、二氢欧山芹醇当归酸酯的线性范围分别为0.2464~2.464μg、0.0228~0.228μg,r均大于0.9991;加样回收率分别为100.85%、101.16%,RSD均小于2.0%。结论不同采收期及生长年限独活中蛇床子素和二氢欧山芹醇当归酸酯的含量差异明显,2~3年生蛇床子素和二氢欧山芹醇当归酸酯的含量逐年增高,3年生春季的含量最大;4~6年的含量逐渐逐年减低,春季采收的独活中两种成分的含量高于秋末采收。
OBJECTIVE To compare the contents of osthol and columbianadin in Angelicae pubescentis Radix from different harvesting time and growing ages. METHODS HPLC method was used as the follow condition: column was Agilent C_(18)( 150 mm × 4. 6 mm,5 μm) and acetonitrile-water( 49∶51) as the mobile phase,detection wavelength was 330 nm,the flow rate was 1. 0 mL·min^(-1),and the column temperature was 30 ℃ . RESULTS The contents of osthol and columbianadin harvested in the early spring of 2-6 years were 0. 39%-1. 11% and 0. 042%-0. 185%,respectively. The contents of osthol and columbianadin were 0. 34%-0. 98% and 0. 034%-0. 177%,respectively,in the late autumn of 2-6 years. The linear range of the mass concentrations of osthol and columbianadin were 0. 2464-2. 464 μg and 0. 0228-0. 228 μg,respectively. The r value of which was all more than 0. 9991. The recovery rates were 100. 8% and 101. 2%,respectively,with the RSD less than 2. 0%.CONCLUSION The contents of osthol and columbianadin from different harvesting time and growing ages are significantly different,which are increased in 2-3 years and decreased in 4-6 years year by year. The contents are highest in the 3-year spring. The contents of the two components in spring harvest are higher than that in late autumn harvest.
