肠道病毒71型诱发的重症手足口病患儿中致病基因表达筛选 被引量:1

摘要 目的:筛选肠道病毒71型诱发的重症手足口病的相关标志物。方法:酶联免疫吸附实验检测重症手足口病患儿外周血中相关细胞因子的表达水平,RNA-Seq检测重症手足口病患儿外周血中单个核细胞的差异基因,Real-time PCR方法验证差异基因的表达情况。结果:重症手足口患儿外周血中细胞因子IFN-γ、TNF-α、IL-4和IL-6表达增高。RNA-Seq筛选出重症手足口患儿差异表达显著的上调基因6个(IFI27、CD177、CD27、TMIGD2、TXNDC5和SIT1)。进一步验证发现IFI27和CD27在重症手足口病中显著增高。结论:差异基因有望成为诊断肠道病毒71型诱发的重症手足口病的分子标记物。 Objective To screen the relevant markers of severe hand,foot and mouth disease(HFMD)induced by enterovirus 71.Methods Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of cytokines in peripheral blood of children with severe HFMD.RNA-seq was used to detect the differentially expressed genes in peripheral blood mononuclear cells of children with severe HFMD.Real-time PCR was used to verify the differential gene expression.Results The expression of cytokines IFN-γ,TNF-α,IL-4 and IL-6 in peripheral blood of children with severe HFMD was increased.RNA-seq screened out 6 significantly up-regulated genes(IFI27,CD177,CD27,TMIGD2,TXNDC5 and SIT1)in severe HFMD children.Further verification showed that IFI27 and CD27 were significantly increased in severe HFMD.Conclusion The differentially expressed genes may be used as molecular markers for the diagnosis of severe hand foot mouth disease induced by enterovirus 71.
