葛根退赤汤对碱烧伤诱导SD大鼠角膜新生血管的抑制作用 被引量:2

摘要 目的研究葛根退赤汤对碱烧伤诱导斯泼累格·多雷(SD)大鼠角膜新生血管(CNV)的抑制作用及其可能作用机制。方法成年SD大鼠40只(40眼),随机分为正常对照组、碱烧伤组、治疗低剂量组、治疗高剂量组。正常对照组10只(10眼)不做处理。剩余30只(30眼)利用1 mol/L NaOH滤纸片建立右眼碱烧伤模型。碱烧伤组予等量生理盐水,治疗组予葛根退赤汤灌胃14 d。于造模成功后第4、8、13天裂隙灯显微镜下观察CNV生长情况,并对角膜炎症反应进行评分。第14天,取角膜组织行苏木精-伊红(HE)染色,观察组织学改变。夹心法(ELISA)检测SD大鼠外周血中血管内皮生长因子(VEGF-A)的表达情况。结果造模后第4、8、13天,治疗低、高剂量组角膜炎症反应评分、CNV的面积均明显小于碱烧伤组,差异有统计学意义(P<0.05)。HE病理染色显示,碱烧伤组角膜组织可见大量炎性细胞浸润及CNV生成,治疗组角膜可见少数CNV。ELISA检测显示,治疗低、高剂量组大鼠外周血中VEGF-A的表达值均明显低于碱烧伤组,且高剂量组表达值低于低剂量组,差异有统计学意义(P<0.05)。结论葛根退赤汤能够减轻碱烧伤后SD大鼠角膜炎症反应,抑制CNV生成,其作用机制可能与减少VEGF-A表达,促进CNV血管内皮细胞凋亡相关。 Objective To research the inhibitory effect of Gegen Tuichi Decoction on corneal neovascularization(CNV)induced by alkali burn in SD rats and its possible mechanism of action.Methods A total of 40 adult SD rats(40 eyes)were randomly divided into the normal control group,the alkali burn group,the low-dose treatment group and the highdose treatment group.Ten rats(10 eyes)in the normal control group were not treated.The remaining 30 rats(30 eyes)were treated with 1 mol/L NaOH filter paper and the right eye alkali burn model was established.The alkali burn group was given the same volume of normal saline,while the treatment group was given Gegen Tuichi Decoction by stomach feeding for 14 days.The growth condition of CNV was observed under slit-lamp microscope on the 4 th,8 th and 13 th day after successful modeling,and corneal inflammatory reaction was scored.On the 14 th day,corneal tissue was stained with hematoxylin and eosin,and histological changes were observed.The expression condition of vascular endothelial growth factor(VEGF-A)in peripheral blood of SD rats was detected by the double antibody sandwich ELISA method.Results On the 4 th,8 th and 13 th day after modeling,the scores of corneal inflammatory reactions and CNV area in the low and high dose groups were significantly smaller than those in the alkali burn group,and the differences were statistically significant(P<0.05).HE pathological staining showed that a large number of inflammatory cells infiltrated and CNV was produced in corneal tissue of the alkali burn group,while a few CNVs were found in corneal tissue of the treatment group.ELISA showed that the expression value of VEGF-A in peripheral blood of rats in the low and high dose groups was significantly lower than that in the alkali burn group,and the expression value of VEGF-A in the high dose group was lower than that in the low dose group,the differences were statistically different(P<0.05).Conclusion Gegen Tuichi Decoction can alleviate corneal inflammatory reaction and inhibit CNV production in SD rats after alkali burn,and its mechanism of action may be related to reducing VEGF-A expression and promoting CNV vascular endothelial cell apoptosis.
