Optimization of extraction technology of total flavonoids component from Guangming eye-protecting gel paste by orthogonal test method

目的优化光明护眼凝胶膏贴剂中总黄酮类成分提取工艺。方法紫外-可见分光光度法测定光明护眼凝胶膏贴剂总黄酮质量分数,选用L9(34)正交表,以乙醇体积分数、乙醇用量、提取时间和提取次数为考察因素,分别以总黄酮提取率为考察指标,优化散痛风凝胶膏贴剂提取工艺参数。结果总黄酮最佳提取工艺A3B1C2D3为最佳提取条件,即采用80%醇提取法,在提取3次,加醇量为6倍量,提取时间1.0 h。结论优化的工艺简单、稳定、可行,重复性好,可分别用于光明护眼贴膏剂中总黄酮的提取。

Objective To optimize the extraction technology of total flavonoids component from Guangming eyeprotecting gel paste.Methods Ultraviolet-visible spectrophotometry was used to determine the total flavonoids component from Guangming eye-protecting gel paste.L9(34)orthogonal table was used to investigate the factors of ethanol volume fraction,ethanol dosage,extraction time and extraction frequencies.The extraction rate of total flavonoids as an index,the extraction technology parameters of the Santongfeng gel paste were optimized.Results The best extraction technology of total flavonoids was A3 B1 C2 D3,which was the best extraction condition,namely,80%alcohol extraction method was adopted,three times more extraction was applied,6 times of standard dosage of ethanol was added,and the extraction time was 1.0 hour.Conclusion The optimized technology is simple,stable,feasible and reproducible,which can be used to extract total flavonoids from Guangming eye-protecting gel paste.
