长枝木霉T6微波诱变菌株筛选及其耐盐性测定 被引量:2

摘要 为了获得高效耐盐木霉菌株,本试验以长枝木霉T6菌株为出发菌株,采用微波诱变处理和NaCl溶液模拟盐胁迫条件,并结合菌丝生长速率,产孢量及菌丝干重等指标筛选最佳诱变耐盐菌株。结果表明,微波诱变对长枝木霉T6菌株生长具有明显影响,并随着微波诱变处理时间的不同其影响作用显著不同,尤其当诱变时间为60 s时,其诱变菌株的菌落直径、菌丝干重和产孢量显著高于原出发菌株。微波诱变时间为60 s条件下,获得的诱变菌株生长速率显著高于其他处理,较原出发菌株表现出较强的耐盐性,培养72 h较原出发菌株生长速率平均提高32.98%。该试验利用微波诱变处理选育和获得了高产和高效的耐盐性木霉菌株,为今后多功能木霉制剂的研发应用及推广奠定理论基础。 The aim is to obtain the high-efficiency salt tolerant Trichoderma strains.The strain of Trichoderma longibrachiatum T6 was used as the original strain.Microwave mutagenesis and NaCl solution were used to simulate the salt stress condition.The colony growth rates, sporulation and mycelium dry weight were considered as indexes to screen the optimal salt tolerant strains.The results showed that the microwave mutation had a significant effect on the growth of Trichoderma longibrachiatum T6, and the effect was different with the different time of microwave mutation.The colony diameter, mycelium dry weight and sporulation of the mutant strain were significantly higher than those of the original strain at 60 s after microwave mutation.In addition, the colony growth rate of the mutant strain was significantly faster than other treatments, and showed stronger salt tolerance than the original strain.The colony growth rate was increased by 32.98% after 72 h of culture compared with the original strain.The high-yield and high-efficiency Trichoderma strains with salt tolerance were selected and obtained in this experiment, which laid a theoretical foundation for the development and application of multifunctional Trichoderma preparation in the future.
