正交实验优化玉液汤煎膏剂中葛根素和总皂苷提取工艺及制备工艺的研究 被引量:2

The Optimization of the Extraction Technology and Preparation Technology of Puerarin and Total Saponins in Yuye Decoction Agent by the Orthogonal Experiment

摘要 优化了玉液汤煎膏剂的制备工艺。以玉液汤的出膏率、总皂苷含量、葛根素含量和醇浸出物作为评价指标,以加水量、浸泡时间、提取次数和提取时间为考察因素,选取单因素试验和正交设计优化处方药材提取工艺;以出膏率、醇浸出物及总皂苷含量作为评价指标,优选出最佳浓缩工艺。经实验得出玉液汤煎膏剂的最佳制备工艺是原方药材回流煎煮两次,分别为加入10倍水量、浸泡80min、回流提取120min和加入8倍水量回流提取90min,将两次药液合并,采用常压90~100℃,或在减压0.07~0.09MPa、温度60~90℃条件下浓缩,相对密度不低于1.22,加入处方量30%的木糖醇,加适量水至1000m L,混匀,灌装,灭菌。该制备工艺合理、稳定可行,适用于玉液汤煎膏剂的工业化生产。 The preparation process of Yuye decoction is optimized. The extraction rate, total saponin content, puerarin content and alcohol extractum of Yuye decoction are used as the evaluation indexes, and the water amount, soaking time, extraction times and extraction time are taken as the investigation factors to perform the single factor test and orthogonal design to optimize the extraction process of prescription medicinal materials;And the optimal concentration process is selected with the extraction rate, alcohol extractum and total saponin content as the evaluation indicators.According to the experiment, the best preparation process of Yuye decoction is that the original herbal medicine is decocted twice with reflux. Ten times amount of water are added, soaks it for 80 minutes, then extracts with the reflux for 120 minutes, and adds 8 times amount of water to extract with the reflux for 90 minutes. Combines the two drug solutions, then perform the concentration process at normal pressure and 90 ~100℃, or at 0.07~0.09 MPa and 60 ~90℃. The relative density is not less than 1.22. The 30% of the prescription amount of xylitol is added and appropriate amount water is added to 1000 mL and mixed well, then fills and make it sterilize. The preparation process is reasonable, stable and feasible, and it is suitable for the industrial production of Yuye decoction.
