不同培养条件对六妹羊肚菌菌丝形态的影响

Effects of Diffferent Culture Conditions on Mycelium Morphology for Morchella sextelata

为考察六妹羊肚菌(Morchella sextelata)菌丝生长特性和羊肚菌液体菌种培养条件,通过单因素试验和正交优化试验,分别研究了平板培养、静置天数、装液体积、玻璃珠、增稠剂等条件对六妹羊肚菌菌丝形态特征、菌丝生物量、菌丝球密度的影响。结果表明,在500 mL基础培养基中添加含0.9%羧甲基纤维素钠的液体培养基300 mL,在25℃黑暗条件下,先静置培养1 d后,150 r·min^(-1)条件培养4 d,其菌丝球均匀,密度3.55×10^(3)个/mL,菌丝生物量最大达8.17 mg·mL^(-1)。此方法有利于羊肚菌菌丝球的形成,且菌球密度高,生物量大,为羊肚菌液体培养提供了技术支撑。

In order to investigate the mycelium growth characteristics and the culture conditions of the liquid spawns culture for M.sextelata,the effects of plate culture,time of static culture,working volume and glass beads on mycelial morphology,mycelium biomass and mycelium pellet concentration were investigated by single factor experiment and orthogonal experiment optimization.The results showed that,300 mL and 0.9%sodium carboxymethyl cellulose was added to each 500 mL basal medium,shock culturing 4 d after static culture 1 d in the dark.Under this condition,the maximum dry weight of mycelium was 8.17 mg·mL-1,and the concentration of mycelium pellets was 3.55×103 per milliliter,and this culture conditions was an effective way for the preparation liquid seed of Morchella spp..
