一测多评法测定印度獐牙菜中8个[口山]酮类成分的含量 被引量:6

Determination of eight xanthones components in Swertia chirayita with quantitative analysis of multi-components by single marker

摘要 目的建立测定印度獐牙菜Swertia chirayita中8个[口山]酮类成分的一测多评测定方法。方法采用高效液相色谱法,以芒果苷为内参物,建立与1,3,5,8-四羟基[口山]酮、swerpunilactone A、1-羟基-3,5-二甲氧基[口山]酮、1-羟基-3,7,8-三甲氧基酮、雏菊叶龙胆[口山]酮、1,8-二羟基-3,5-二甲氧基[口山]酮和1,8-二羟基-3,7-二甲氧基[口山]酮的相对校正因子(f_(s/i)),并利用f_(s/i)计算印度獐牙菜中各成分的含量,同时采用外标法测定该8个成分的含量,比较两者的差异,以验证一测多评法的可行性与准确性。结果各校正因子的重复性良好,在12批印度獐牙菜中运用一测多评法和外标法得到的各成分含量无显著性差异(RSD<3%)。结论建立的一测多评法准确性高,可应用于印度獐牙菜的含量测定和质量评价。 Objective To establish a quantitative analysis of multi-components by single marker method for the determination of eight xanthones in Swertia chirayita.Methods The relative correction factors(f_(s/i))of seven xanthones(1,3,5,8-tetrahydroxyxanthone,swerpunilactone A,1-hydroxy-3,5-dimethoxyxanthone,1-hydroxy-3,7,8-trimethoxyxanthone,bellidifolin,1,8-dihydroxy-3,5-dimethoxyxanthone and 1,8-dihydroxy-3,7-dimethoxyxanthone)were determined by HPLC method with the mangiferin as the internal standard,which were used to calculate the content of each component in S.chirayita.At the same time,the external standard method was used to determine the content of the eight components,and the differences between the two methods were compared to verify the feasibility and accuracy of quantitative analysis of multi-components by single marker.Results The reproducibility of each correction factor was good,and there were no significant differences in the contents of the ingredients obtained by using the quantitative analysis of multi-components by single marker and the external standard method in the 12 batches of S.chirayita(RSD<3%).Conclusion The established quantitative analysis of multi-components by single marker has high accuracy and can be applied to the content determination and quality evaluation of S.chirayita.
