微小RNA Let-7在子宫肌瘤组织中的表达情况及与复发相关危险因素的探讨

The expression of microRNA Let-7 in uterine fibroids and the risk factors related to recurrence

目的:探讨单发、多发子宫肌瘤、不同直径子宫肌瘤以及肌瘤与相应肌壁组织中miRNA Let-7的表达情况,探讨miRNA Let-7与子宫肌瘤复发危险因素的关系。方法:运用实时定量聚合酶链反应(real-time,PCR)方法分别检测单发肌瘤与多发子宫肌瘤、不同直径的肌瘤组织以及对应肌壁组织中miRNA Let-7的相对表达量,并对分组数据进行差异分析。结果:子宫肌瘤组织中的miRNA Let-7相对表达量略低于肌瘤旁对应肌层组织,差异无统计学意义(P>0.05);单发肌瘤病例组织中的相对表达量明显高于多发肌瘤组,差异有统计学意义(P<0.05);肌瘤直径<5 cm组病例中的相对表达量明显高于≥5 cm组,差异有统计学意义(P<0.05)。结论:子宫肌瘤的数目、直径大小是术后复发的独立危险因素,miRNA Let-7在多发肌瘤、大直径肌瘤病例中的相对高表达可以为子宫肌瘤复发机制的研究提供新的理论依据。

Objective:To investigate the expression levels of microRNA Let-7 in tissues of uterine myoma and surrounding normal myometriumin,all cases were classified into different groups according to the number or diameter of myomas,then analyze the relations between microRNA Let-7 and risk factors of myoma recurrence.Methods:The microRNA let-7 expression levels in uterine myoma and surrounding normal myometrium in tissues were detected by quantitative real-time PCR from groups of single or multiple uterine myoma,larger myomas(diameter≥5 cm) or smaller myomas(diameter<5 cm).Results:The expression levels of microRNA let-7 in myoma tissues were slightly weaker than that of surrounding normal myometriumin,but the difference was not significant(P>0.05).Then the expression levels of microRNA let-7 in groups of single myoma and smaller myoma(diameter<5 cm) were higher than of multiple myoma and larger myoma(diameter≥5 cm),and the difference was statistically significant(P<0.01).Conclusion:The number and diameter of myomas are independently risks of recurrence.MicroRNA Let-7 with higher expression in multiple myoma and larger myoma is expected to be an new marker for myoma recurrence.
