摘要
目的:探究脱氧核苷酸钠对结肠癌细胞化疗敏感性及白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、Survivin表达的影响。方法:将结肠癌细胞分为6组:对照组、奥沙利铂组(100μg·L^(-1))、脱氧核苷酸钠组(100μmol·L^(-1))、脱氧核苷酸钠低、中、高剂量(20,60,100μmol·L^(-1))+奥沙利铂(100μg·L^(-1))组。采用MTT实验和流式细胞仪检测SW620细胞增殖和凋亡情况,实时荧光定量PCR(qRT-PCR)检测SW620细胞中IL-6、IL-8、Survivin mRNA表达情况,蛋白免疫印迹(Western blot)检测SW620细胞中IL-6、IL-8、Survivin蛋白表达情况。结果:脱氧核苷酸钠组SW620细胞增殖率、凋亡率、IL-6、IL-8、Survivin mRNA及蛋白表达水平同对照组相比较,差异均无统计学意义(P>0.05)。奥沙利铂组和脱氧核苷酸钠+奥沙利铂各组SW620细胞增殖率显著低于对照组(P<0.05),脱氧核苷酸钠中、高剂量+奥沙利铂组SW620细胞增殖率显著低于奥沙利铂组和脱氧核苷酸钠低剂量+奥沙利铂组(P<0.05);奥沙利铂组和脱氧核苷酸钠+奥沙利铂组SW620细胞凋亡率显著高于对照组(P<0.05),脱氧核苷酸钠中、高剂量+奥沙利铂组SW620细胞凋亡率显著高于奥沙利铂组和脱氧核苷酸钠低剂量+奥沙利铂组(P<0.05);奥沙利铂组和脱氧核苷酸钠+奥沙利铂组SW620细胞中IL-6、IL-8、Survivin mRNA及蛋白表达水平显著低于对照组(P<0.05),脱氧核苷酸钠中、高剂量组SW620细胞中IL-6、IL-8、Survivin mRNA及蛋白表达水平显著低于奥沙利铂组和脱氧核苷酸钠低剂量+奥沙利铂组(P<0.05),且呈剂量依赖性。结论:脱氧核苷酸钠可能通过下调结肠癌细胞中IL-6、IL-8、Survivin表达,增加结肠癌细胞化疗敏感性。
Objective: To investigate the effects of deoxyribonucleotide sodium on the chemosensitivity of colon cancer cells and the expressions of interleukin-6( IL-6),interleukin-8( IL-8) and Survivin. Methods: The colon cancer cells were divided into 6 groups: control group,oxaliplatin group( 100 μg·L^(-1)),deoxyribonucleotide sodium group( 100 μg·L^(-1)),and low,medium and high dose deoxyribonucleotide sodium( 20,60,100 μmol·L^(-1)) + oxaliplatin( 100 μg·L^(-1)) groups. MTT assay and flow cytometry were used to detect the proliferation and apoptosis of SW620 cells,real-time fluorescence quantitative PCR( qRT-PCR) was used to detect the expressions of IL-6,IL-8 and Survivin mRNA in SW620 cells,and the expressions of IL-6,IL-8 and Survivin in SW620 cells were detected by protein immunoblotting( Western blot). Results: The proliferation rate,apoptosis rate,and IL-6,IL-8,Survivin mRNA and protein expression levels of SW620 cells in deoxynucleotide sodium group were not significantly different from those in the control group( P> 0.05). The proliferation rate of SW620 cells in oxaliplatin group and deoxyribonucleotide sodium + oxaliplatin groups was significantly lower than that in the control group( P<0.05). The proliferation rate of SW620 cells in middle and high dose deoxyribonucleotide sodium + oxaliplatin groups was significantly lower than that in oxaliplatin group and low dose deoxyribonucleotide sodium + oxaliplatin group( P<0.05). The apoptosis rate of SW620 cells in oxaliplatin group and deoxyribonucleotide sodium + oxaliplatin groups was significantly higher than that in the control group( P<0.05). The apoptosis rate of SW620 cells in middle and high dose deoxyribonucleotide sodium + oxaliplatin groups was significantly higher than that of oxaliplatin group and low dose deoxyribonucleotide sodium + oxaliplatin group( P<0.05). The mRNA and protein expression levels of IL-6,IL-8 and Survivin in SW620 cells of oxaliplatin group and deoxyribonucleotide sodium + oxaliplatin groups were significantly lower than those of the control group( P <0.05). The expression levels of IL-6,IL-8 and Survivin in SW620 cells of middle and high dose deoxyribonucleotide sodium + oxaliplatin groups were significantly lower than those in oxaliplatin group and low dose deoxyribonucleotide sodium + oxaliplatin group( P <0.05) in a dose-dependent manner. Conclusion: Deoxyribonucleotide sodium may increase the chemosensitivity of colon cancer cells by down-regulating the expressions of IL-6,IL-8 and Survivin in colon cancer cells.
