Box-Behnken效应面法优化微波辅助提取都梁软胶囊中4种有效成分的工艺

Optimization of Microwave-assisted Extraction for 4 Active Constituents from Duliang Soft Capsules by Box-Benhken Response Surface Method

目的:采用Box-Behnken效应面法优选都梁软胶囊中欧前胡素、异欧前胡素和藁本内酯的微波辅助提取工艺条件。方法:在单因素试验的基础上,采用四因素三水平Box-Behnken试验设计对都梁软胶囊中欧前胡素、异欧前胡素和藁本内酯的微波辅助提取工艺参数进行优选。以乙醇浓度、料液比、提取时间及微波功率为自变量,采用Hassan方法计算欧前胡素、异欧前胡素和藁本内酯的总评"归一值",建立总评归一值与自变量之间的数学关系,经效应面法预测最佳工艺条件。结果:最佳提取工艺为乙醇浓度59%、料液比44 ml·g^(-1)、提取时间8.2 min、微波功率600 W;欧前胡素、异欧前胡素及藁本内酯的含量预测值分别为2.132 0,1.241 0,1.840 0 mg·g^(-1)。结论:实测值与预测值接近,表明采用Box-Behnken效应面法优化后得到的微波辅助提取工艺参数可用于都梁软胶囊中欧前胡素、异欧前胡素和藁本内酯的提取。

Objective: To optimize the processing parameters of the microwave-assisted extraction(MAE) of imperatorin,isoimperatorin and ligustilide in Duliang soft capsules by Box-Behnken experimental design. Methods: On the basis of single factor screening,a four-factor and three-level Box-Behnken experimental design was employed with ethanol concentration,liquid ratio,extraction time and microwave power as independent variables. Dependent variables were transformed into desirabilities mathematically of imperatorin,isoimperatorin and ligustilide by Hassan’s method. The mathematical relationship was established between the dependent variables and the independent variables. Optimum experimental conditions were selected from the stationary point of response surfaces. Results: The optimum processing conditions were as follows: the materials were extracted for 8.2 min with 44-fold amount of 59% ethanol,and the microwave power was 600 W. The content of imperatorin,isoimperatorin and ligustilide was 2.132 0,1.241 0 and 1.840 0 mg·g^(-1),respectively. Conclusion: The measured values are close to the predicted values,which indicates the comprehensive microwave-assisted extraction parameters optimized by Box-Behnken experimental design can be used for the extraction of imperatorin,isoimperatorin and ligustilide from Duliang soft capsules.
